Abstract

Objective To investigate the effect of Toll like receptor 2 (TLR2) on Dectin-1 expression in HBE cells resulted from Aspergillus fumigatus stimulation.Methods The infection model in vitro was established via HBE cells exposed to Aspergillus fumigatus,and the mRNA and protein expressions of Dectin-1 and TLR2 were respectively detected by PCR and Westen blot.To explore the effect,TLR2 was silenced by siRNA and the infection model was copied.Then,the total expression of Dectin-1 was detected by Western blot and its expression on cell membrance was observed through the flow cytometry.Results TLR2 expression in HBE cells demonstrated an increasing trend after Aspergillus fumigatus stimulation,and reached a peak after 18 hours.However,the expression was not significantly different from the basal expression.Notably,the expression of Dectin-1 both in whole cell and on cell membrance was inhibited after 18 hours when TLR2 was silenced.Conclusions Our findings reveal an increasing expression of TLR2 after HBE cells exposed to Aspergillus fumigatus.And the expression of Dectin 1 in HBE cells triggered is probablely based on TLR2's early recoganization of Aspergillus fumigatus which activates the downstream signal transduction pathyway.
