Abstract

目的 探讨肉苁蓉总苷(GCs)对D-半乳糖(D-gal)脑老化模型小鼠的保护作用机制.方法实验分30d和50d,每期NIH小鼠各为120只,随机分为6组:正常对照组(NC)、模型组(D-gal)、VitaminE组、GCs组:分小、中、大剂量组[(mg.kg -1.d -1)31.25、62.5、125],每组20只.除NC组外,在各组小鼠皮下注射D-gal(50 mg.kg -1.d -1 );NC组皮下注射NS, D-gal组和NC组给予等容积饮水灌胃,VitaminE组和GCs组灌胃给药,每日1次,均连续皮下注射和灌胃30d和50d.分别测定5min错误次数、潜伏期、脑组织SOD的活性、MDA、LF含量,并观察50d后海马CA1区组织结构形态的变化.结果 (1)5min内的错误次数: 30d和50d的D-gal组分别为(8.25±4.03)次和(9.63±6.18)次,均显著高于其它各组(P 均< 0.01).(2)潜伏期: 30d和50d的D-gal组分别为 (25.21±13.86)s和(31.75± 17.77)s,低于NC组、GCs中剂量组和大剂量组(P <0.05,P <0.01).(3)SOD活性: 30d和50d D-gal组脑组织的SOD活性较其它各组低(P <0.05,P <0.01).(4)MDA含量:30d和50d D-gal组脑组织的MDA含量均较其它各组高(P <0.05,P <0.01).(5)LF含量:30d和50d D-gal组脑组织的LF含量均较NC组高,30d的脑组织GCs小剂量组、VitaminE组及50d GCs各组及VitaminE组均较D-gal组低(P <0.05,P <0.01).(6)50d的D-gal组小鼠海马CA1区与NC组、GCs大剂量组及VitaminE组的组织形态不同.结论 GCs能显著改善不同天数模型小鼠的学习、记忆功能,升高脑组织的SOD活性,降低MDA、LF含量,并对海马CA1区组织结构形态学改变有保护作用.提示GCs可能是通过抗氧化的机制达到改善脑老化的作用。
