Abstract
A complete amino acid sequence containing 210 residues of a rabbit light chain of a homogeneous Group C streptococcal antibody has been determined as: See journal for formula. This light chain is allotype b4 and is of the V-K1 subgroup.
Highlights
Further sequence analyses of peptides from tryptic and peptic digests have allowed assignment of the complete amino acid sequence of 210 residues for this light chain. Comparison of this sequence to other available sequences reveals a high degree of homology to human V,l light chains
A tentative identification of hypervariable positions can be made by comparison of the V region sequence of this antistreptococcal antibody with those of light chains obtained from rabbit antibodies sequenced in other laboratories
After digestion for 2 hours at 37”, the digest was directly applied to a Sephadex G-50 column (3 X 200 cm) which was pre-equilibrated with 0.02 M NHdOH for peptide separation. (b) Trypsin digest: 100 mg of the completely reduced and carboxamidomethylated L chain were suspended in 20 ml of 0.2 M NH,HCOs, 1 mg of lyophiliaed trypsin was added
Summary
A complete amino acid sequence containing 210 residues of a rabbit light chain of a homogeneous Group C streptococcal antibody has been determined as: 10. In the previous paper of this series, results on the partial sequence determination of a rabbit light chain of allotype b4 were described (1). Further sequence analyses of peptides from tryptic and peptic digests have allowed assignment of the complete amino acid sequence of 210 residues for this light chain. Comparison of this sequence to other available sequences reveals a high degree of homology to human V,l light chains. A tentative identification of hypervariable positions can be made by comparison of the V region sequence of this antistreptococcal antibody with those of light chains obtained from rabbit antibodies sequenced in other laboratories
