Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian glukosa dan inhibitor glikolisis terhadap viabilitas sperma domba lokal. Penelitian ini dilaksanakan pada bulan Januari - Mei 2024 dengan objek penelitian berupa semen dari tiga pejantan domba lokal yang berumur 2-3 tahun. Metode Rancangan Acak Lengkap (RAL) digunakan pada penelitian ini dengan 4 perlakuan yaitu tanpa pemberian glukosa (P0), pemberian glukosa 27 mM (P1), 54 mM (P2), dan 108 mM (P3) dengan inkubasi pada 37°C selama 0-60 menit. Setiap perlakuan diulangi sebanyak 5 kali. Kemudian setelah perlakuan optimal pada eksperimen pertama diketahui, dilanjutkan dengan eksperimen kedua dengan pemberian 3-MCPD 0, 0,5, 1, dan 2 mM pada media bufer yang mengandung glukosa 27 mM. Setiap perlakuan diulang 5 kali. Data hasil penelitian dianalisis menggunakan sidik ragam dan dilanjutkan dengan uji Tukey HSD. Hasil penelitian menunjukkan bahwa pemberian glukosa pada dosis tertentu secara signifikan mempertahankan viabilitas sperma. Pemberian glukosa 27 mM (P1) nyata mampu mempertahankan viabilitas lebih tinggi yaitu 75 ± 2,7%, dibanding P0 (67 ± 2,4%), P2 (74 ± 0,71%), dan P3 (72 ± 0,69%) yang berarti bahwa sperma menggunakan glukosa untuk mempertahankan integritas membrane. Namun, pengaruh glukosa dalam mempertahankan viabilitas sperma terhambat dengan kehadiran 3-MCPD (0,5 mM, 65,8 ± 1%; 1 mM, 61,6 ± 1,2%; dan 2 mM, 55,4 ± 2,2%). Kesimpulan dari penelitian ini yaitu pemberian glukosa dapat mempertahankan viabilitas sperma kambing melalui proses glikolisis.The purpose of this research was to determine the effect of glucose supplementation and 3-MCPD (a glycolysis inhibitor) on sperm viability of Local Ram. This research was conducted from January - May 2024 with the object of research was sperm from 2 to 3-year-old male Local Ram. The research applied Completely Randomized Design (CRD) with four treatments i.e.: sperm without glucose supplementation (P0), 27 mM glucose (P1), 54 mM glucose (P2), and 108 mM glucose (P3) with incubation at 37°C for 0-60 min. Each treatment was replicated five times. Then after the optimal treatment in the first experiment was known, continued with the second experiment by adding 0, 0.5, 1, or 2 mM 3-MCPD in a buffer medium containing 27 mM glucose. Each treatment was replicated five times. The obtained data were analyzed using the ANOVA and Tukey HSD test. The results showed that addition of glucose at certain doses significantly maintained the viability of sperm. The addition of 27 mM glucose (P1) has the highest sperm viability by 75 ± 2.7%, compared to P0 (67 ± 2.4%), P2 (74 ± 0.71%), and P3 (72 ± 0.69%), suggesting sperm utilise glucose to support their membrane integrity. However, the supporting effect of glucose on sperm viability was abolished by the presence of 3-MCPD (0.5 mM, 65.8 ± 1%; 1 mM, 61.6 ± 1.2%; and 2 mM, 55.4 ± 2.2%). In conclusion, ram sperm utilise glucose to maintain their viability via glycolysis.
