Abstract

A technical procedure is described for the enzymatic isolation of the corn embryo sac. Light and fluorescence microscopy were used to assess the condition of the embryo sac. The results show that all of the constituent cells of the isolated corn embryo sac are structurally intact, viable easily identified, and contain only remnants of a cell wall. This procedure is a pre-requisite for the successful preparation of isolated female gametes.
