Abstract

Genetic diversities of nuclear and chloroplast genomes were studied in the wild einkorn wheat, Triticum urartu, and compared with those in the other einkorn wheat species, T. boeoticum and T. monococcum, and two diploid Aegilops species, Ae. speltoides and Ae. squarrosa. Amplified fragment length polymorphism (AFLP) and simple sequence length polymorphism (SSLP) analyses were employed. Measurement of interspecific genetic distances and cluster analysis of 55 accessions of the einkorn wheat clearly showed that T. urartu was greatly differentiated from the other two A genome species. A similar result was obtained by the chloroplast SSLP analysis. The intraspecific DNA variations of nuclear genomes within the einkorn wheat were smaller than those within the two Aegilops species. In contrast, the einkorn wheat showed larger chloroplast DNA variations than the others. Ae. speltoides showed the largest nuclear genome variation, while its chloroplast genome variation was the least.
