Abstract

Aging is a major risk factor for cardiac morbidity and mortality. Previously, we showed that cardiac aging is associated with a decreased E/A ratio, which was preserved in the matrix metalloproteinase‐9 (MMP‐9) null mice. To evaluate plasma changes that occur before cardiac dysfunction, we examined 15–18 month old wild type (WT, n=16) and MMP‐9 null (null, n=21) mice. By Doppler, the E/A ratios were similar between the 2 groups (1.42±0.05 in WT vs. 1.38±0.06 in null). Of 60 analytes evaluated in plasma, 2 were lower and 6 were higher in the null group. The percent differences were −20% for CD40, −46% for haptoglobin, 129% for immunoglobulin A, 35% for lymphotactin, 13% for macrophage colony stimulating factor−1, 28% for stem cell factor, 16% for vascular cell adhesion molecule‐1, and 50% for von Willebrand factor (all p<0.05). These results suggest that MMP‐9 deletion decreases age‐associated inflammation before cardiac function is impaired.
