Abstract

A lectin corona on the surface of Fe3O4 magnetic nanoparticles (MNP) that can specifically co-localize glycoenzymes with enhanced stability and catalytic performance is constructed. Minimal conformational distortion of enzymes is achieved with ConA corona coated MNP, as minor shifts in circular dichroism (CD) spectrum are observed compared to that without ConA. In comparison to the freely diffusing enzymes, the GOx-CAT enzyme cascades immobilized on ConA-MNP results in 1.3 fold enhanced activity, which can be attributed to the nanoscale proximate packing of cascade glycoenzymes. The affirmed stabilization and activation effect, in addition to the ease of recycle and operation assured by MNP, offer new possibilities for the ConA-MNP-carried enzymatic cascades.
