Juniperus oxycedrus, from the Cupressaceae family, is widespread in the Mediterranean. However, the allergenic relevance of its pollen remains underexplored. Objective: This study aimed to characterize Jun o 1, an allergen of J oxycedrus that is homologous to major allergens in related species. Native Jun o 1, its nonglycosylated Escherichia coli-expressed form (rJun o 1), and Cup a 1 were purified by affinity chromatography. Jun o 1 was characterized by SDS-PAGE, 2D-electrophoresis, Western blot (WB), and mass spectrometry and compared with the homologous cypress allergen, Cup a 1. Enzymatically and chemically deglycosylated Jun o 1 was also analyzed. Specific IgE binding and cross-reactivity were evaluated using ELISA, WB, and mast cell activation assays with sera from 57 Spanish cypress-allergic patients. IgG binding was tested using sera from rJun o 1-immunized mice. Jun o 1 exists as multiple isoforms and assembles into oligomers, whereas Cup a 1 comprises multiple isoforms without oligomerization. IgE binding to Jun o 1, including mast cell activation, is critically influenced by the combined presence of protein and carbohydrate determinants. Compared to Cup a 1, Jun o 1 exhibited greater IgE-binding inhibition for J oxycedrus extract in sera from Cupressaceae-sensitized allergic patients from this tree's autochthonous regions. The highest rate of monosensitization in this group was to J oxycedrus. Jun o 1 is a relevant and potential major allergen in cypress allergy. Structural and immunogenic similarities with Cup a 1 lead to cross-reactivity, although it also exhibits unique properties, resulting from combined IgE-reactive protein and glycan determinants.

Use of IgE and IgG4 epitope binding to predict the outcome of oral immunotherapy in cow's milk allergy

Oral immunotherapy (OIT) with cow's milk (CM) has been reported to induce a number of specific antibody responses, but these remain to be fully characterized. Our objective was to explore whether IgE and IgG4 epitope binding profiles could predict the risk of side effects during CM OIT. The study population consisted of 32 children (6-17 yr of age) with CM allergy: 26 children who successfully completed OIT and six children who discontinued therapy due to adverse reactions. We investigated sera drawn before and after OIT. We analyzed specific IgE and IgG4 binding to CM protein-derived peptides with a microarray-based immunoassay. Antibody binding affinity was analyzed with a competition assay where CM proteins in solution competed with peptides printed on the microarray. IgE binding to CM peptides decreased and IgG4 binding increased following the OIT in children who attained desensitization. Compared with children who successfully completed OIT, those who discontinued OIT due to adverse reactions developed increased quantities and affinity of epitope-specific IgE antibodies and a broader diversity of IgE and IgG4 binding, but less overlap in IgE and IgG4 binding to CM peptides. Detailed analysis of IgE and IgG4 binding to CM peptides may help in predicting whether CM OIT will be tolerated successfully. It may thus improve the safety of the therapy.

Alphas1-Casein (CAS1_BOVIN), the major allergen of cow's milk (CM), is widely used as hydrolysates in infant diet formulae and additive to other processed food items. To date, most of the reported B-cell epitope mapping were performed on polyethylene pins or cellulose-derivative membrane. We sought to locate the motifs critical for human-specific IgE and rabbit polyclonal IgG binding using extensively purified CAS1_BOVIN, synthetic peptides and derivatives. Thirteen overlapping peptides covering the whole CAS1_BOVIN encompassing 17 : 20 amino acid (AA) were synthesized by f-moc AA solid-phase polyamide peptide synthesis. In addition, six cyanogen bromide (CNBr) cleavage fragments were prepared. Limited hydrolysis, oxidized and reduced/alkylated derivatives were also produced. The preparations were purified by ion exchange, gel filtration chromatography, reversed phase and high-performance liquid chromatography. The homogeneity was visualized by sodium dodecyl sulfate (SDS) and poly acryl amide gel electrophoresis (PAGE) followed by IgE and IgG immunoblotting. IgE binding was measured by Biotin Streptavidin (Bio/strep) fluoro enzyme immuno assay (FEIA) or ELISA-inhibition. Eighteen CM allergy (CMA) sera from 45 clinically examined children (Melbourne) and five adults (Bergen) were selected. Individual sera and pools were used for mapping IgE-binding epitopes. Rabbit IgG sera and pools were used for locating the antigenic sites of the molecule. Results indicated that all the individual CMA sera and pools recognized the intact molecule and three of the CNBr fragments as major antibody-binding allergens. The N- and C-terminal peptides (CAS 16-35; CAS 136-155) showed high IgE-binding affinity. CAS 1-18 and CAS 181-199 showed high IgG bindings. Considering the diversity of the antibody specificities, a reasonable agreement between IgE and IgG epitopes were found at the N- and C-terminals of CAS1_BOVIN. Mapping IgE B-cell epitopes by direct Bio/strep FEIA allowed the development of a sensitive modified technique for detecting unlabelled, casein immune dominant peptides in food products.

Predicting Tolerance to Cow's Milk Allergy in Children Using IgE and IgG4 Peptide Binding Profiles.

Cow's milk allergy (CMA) is the most common food allergy in infants. This study aimed to identify peptide biomarkers predictive of tolerance in a Spanish population of children with CMA. We investigated specific IgE and IgG4 binding to sequential epitopes of the five major CM allergens (α-s1-, α-s2-, β-, and κ-caseins as well as β-lactoglobulin) using a microarray-based immunoassay. Microarray analysis was performed in 118 patients at baseline and after 6, 18, 30, 42, and 54 months. Most patients tolerated CM at 6 months (40.7%) and 18 months (35.4%). We found significant differences in IgE and IgG4 binding intensity and diversity between allergic and tolerant patients. No differences were observed at baseline. Combining baseline IgE and IgG4 serology variables and peptide microarray analysis results, a predictive model was developed using the XGBoost algorithm to classify tolerance status at different time points. The generated models showed high predictive value at 6 and 30 months with AUCs of 0.883 and 0.833, respectively. Therefore, using IgE and IgG4 antibody-binding peptides at baseline, we generated two models predicting tolerance in children with cow's milk allergy at 6 and 30 months.

BackgroundOral immunotherapy (OIT) with peanut (Arachis hypogaea) allergen powder-dnfp (PTAH; Aimmune Therapeutics) is an FDA-approved treatment to desensitize peanut allergic participants.ObjectiveHere we assessed shifts in IgE and IgG4 binding to peanut allergens and their epitopes recognized by United States (US) peanut allergic participants (n = 20) enrolled in phase 3 PTAH OIT clinical trials.MethodsPre- and post- trial participant sera were collected approximately 12 months apart and tested for IgE binding to intact peanut proteins via ImmunoCAP ISAC immunoassays. IgE and IgG4 linear epitopes were identified based on binding to synthetic overlapping 15-mer linear peptides of 10 peanut allergens (Ara h 1-11) synthesized on microarray slides.ResultsStatistically significant decreases in IgE binding were identified for intact Ara h 2, 3, and 6, and known and newly identified IgE epitopes were shown to exhibit shifts towards IgG4 binding post-OIT, with most linear peptides having increased IgG4 binding after treatment with PTAH. While PTAH does not seem to alter the actual peptide binding patterns significantly after one year of treatment, the IgE and IgG4 binding ratios and intensity are altered.ConclusionAt a population level, the linear IgE and IgG4 epitopes of 10 peanut allergens overlap and that increase in IgG4 with OIT results in displacement of IgE binding to both conformational and linear epitopes. Furthermore, it appears as though the increase in IgG4 is more important to achieve desensitization at the 12-month timepoint than the decrease in IgE. This type of knowledge can be useful in the identification of IgE and IgG4-binding allergen and peptide biomarkers that may indicate desensitization or sustained unresponsiveness of allergic individuals to peanut.
