Abstract

AIM:To identify the main metabolites of rotundine in rat biliary samples.METHOD:Bile samples of rats after an oral administration of rotundine(120 mg·kg-1) were analyzed by HPLC-MS/MS.The separation was achieved on a Zorbax SB C18 column(150 mm × 0.5 mm I.D., 5μm);mobile phase consisted of methanol-water-triethylamine(30:70:0.07, V/V/V).Positive ESI mode was operated.RESULT:Four metabolites were observed in rat biliary samples, and the structures of two metabolites were tentatively identified as sulfuric acid conjugates of rotundine after demethylation.CONCLUSION:Conjugation with sulfuric acid after demethylation may be one of the main excretion pathways of rotundine in rat bile.
