Abstract

Hyperthermia (HT) has proven to be able to alter invasion ability of cancer cells. However, the detailed mechanisms responsible for the anti‐metastasis effects of HT have not been elucidated. N‐myc downstream‐regulated gene 2 (NDRG2) is a member of NDRG family, which has been suggested to be highly responsive to various stress and associated with tumor suppression. The present study aimed to investigate biological role of NDRG2 in invasion of HCC cells under hyperthermia. We found that NDRG2 could be induced by hyperthermia at 45□. Meanwhile, NDRG2 overexpression can inhibit expression of MMP2 and MMP9 as well as invasion of HCC cells. Whereas knocking‐down of NDRG2 increased invasion ability of HepG2 cells. Moreover, NDRG2 was proved to decline HT‐induced phosphorylation of ERK1/2 notably in HepG‐2 cells exposed to hyperthermia. Taken together, these data suggest that NDRG2 could be induced by hyperthermia and mediates hyperthermia‐caused inhibition of invasion in HCC cells via Suppressing ERK1/2 Signaling Pathway.
