Abstract

A rosette-type assay of the physical association between peripheral blood monocytes and human lymphoblastoid (LB) cell lines is reported. Monocytes formed rosettes with LB B-cell lines at 37°C, but not with T-cell lines. Binding was divalent cation dependent, inhibited by trypsinization, and enhanced by neuraminidase treatment. It was not inhibited by anti-immunoglobulins, heat-aggregated γ-globulin, or corticosteroids. The assay provides a simple means of studying the ability of monocytes to establish cell-to-cell contact with activated lymphocytes.
