Abstract

Background: Fat digestion is reported as subnormal in some patients with pancreatic exocrine insufficiency (P.I.), despite supplementation with high doses of pancreatic enzymes. This problem is being investigated using a minipig model of P.I.. Methods: Studies were made in 6 female (25 -30kg) GOttingen minipigs (Ellegaard) fitted under halothane anaesthesia with an lien-caeca/, re-entrant cannula; 3 of the pigs were made P.L by ligating the pancreatic duct. All pigs had free access to water, and were fed twice daily with 250g of a 30% crude fat (soya oil) diet mixed with 500 ml HzO, and 0.625g CrzO3; the P.I. pigs were given 0, 8, 16 or 24 capsules of pancrelipase (10,000 lipase, 33,200 amylase, 37,500 protease USP units/capsule) mixed with each meal. In each case, after >__ 10 days adaptation, the pigs were put in modified metabolism cages. All stools were collected for 5 days and stored at -20°C. On 3 subsequent days, ileal chyme was collected on ice for 2h periods over 12h and stored at -20°C. Frozen samples were freeze-dried and analyzed: chrome was oxidized to chromate and measured at 365nm (spectrophotometer); crude fat was estimated after acid hydrolysis and petrol ether extraction (gravimetric); dry matter (DM) by freeze drying; digestibility (%) = (g/day in feed g/day in ileum or stool)/(g/day in feed) x 100.
