Abstract

Dako's HercepTest is the only Food and Drug Administration (FDA) approved assay for HER2 oncoprotein overexpression. For the HercepTest to perform as intended, test conditions must conform exactly to the manufacturer's directions. The HercepTest protocol requires heat induced epitope retrieval (HIER) of tissue sections in a 95°C water bath for 40 min. This requirement poses a unique problem for laboratories located at altitudes above 5000 ft, where water boils below 95°C. Rapid boiling can cause tissue sections to be sheared from the surface of a glass slide. This paper will address HEIR methods that can be used at high altitudes to ensure good HER2 immunolabeling.
