Abstract
Propionate- 14C and glyoxylate, in the presence of ATP and CoA, were incubated with cell-free extract of Bacillus sp. grown in propionate medium. The radioactive compound formed was proved to be β-methylmalate from the following evidence: Its R f value in paper chromatography agreed with that of authentic β-methylmalate, and its retention time in gas-liquid chromatography was the same as that of β-methylmalate. After the radioactive compound was mixed with unlabeled β-methylmalate and urea, the mixture was converted to thymine. The specific radioactivity of thymine remained constant through three recrystallization procedures.
