Abstract

The effect of spore concentration, incubation period, culture age, buffer concentration, pH and incubation temperature on germination of Cochliobolus sativus over host leaves has been investigated. The data revealed that for optimum germination and germ tube growth of C. sativus over cleared host leaves, the drops of spore suspension (105 spores per ml), prepared in phosphate buffer (0.01 M, pH 6) using spores collected from 20-day old cultures, should be incubated at 28 ± 1°C for 16 h.
