Abstract

Objective To investigate the Dicer gene expression and its clinicopathological significance in papillary thyroid carcinoma,and the relationship between the Dicer gene expression and tumoro-genesis of papillary thyroid carcinoma.Methods The immunohistochemical streptavidin avidin-peroxidase (SP) technique and real-time quantitative polymerase chain reaction (Real-time PCR) were used to detect the expression of Dicer protein and mRNA in 50 cases of PTC and their matched adjacent non-cancerous epithelia (NCE).Results The positive expression rate of Dicer mRNA in PTC and NCE wase 80%(40/50) and 10% (5/50) respectively (P < 0.01).The positive expression rate of Dicer protein in PTC and NCE was 76% (38/50) and 8% (4/50) respectively (P < 0.01).The expression of Dicer protein and mRNA was associated with the invasion and metastasis of PTC (P < 0.05).The expression of Dicer protein and mRNA showed a distinct correlation (P < 0.01).Conclusion Dicer plays a pivotal role in the carcinogenesis and progression of PTC.Dicer could be a new diagnostic/therapeutic target in PTC.
