Abstract

<h3>Purpose of This Experiment</h3> The purpose of this experiment was twofold: (1) to determine the most effective time of administration of local chemotherapy in experimental Pseudomonas middle ear inflammation; (2) to compare pathological changes in the middle and inner ear with and without local chemotherapy. <h3>Bacteriology</h3> One strain of β-hemolytic Streptococcus, and 2 strains of Pseudomonas aeruginosa were used. These strains were isolated from recent human cases of acute and chronic middle ear suppuration. A 16-hour broth culture of each strain was used for inoculation. No bacterial emulsion was used. <h3>Animals</h3> Young male guinea pigs, 1-1¼ lb., were acclimated to animal room feed and environment for several days. Only animals which had no evidence of middle ear infection were used. <h3>Inoculation</h3> To obtain general anesthesia, 0.25-0.3 cc. of pentobarbital (Sodium Nembutal) 50 mg/cc was injected intraperitoneally.
