Estimation of phytochemicals, antioxidant, antimicrobial, and brine shrimp lethality activities of Valeriana jatamansi Jones

Herbal medicine has been widely utilized in traditional medicine for treating various ailments. This study aimed to quantify the phenolic, flavonoid, and tannin content in different plant extracts, assess their antioxidant capacity, and evaluate their antimicrobial efficacy. Among the extracts tested, methanol exhibited the highest total phenolic content (TPC) at 166.5 ± 2.0 mg GAE/g and total flavonoid content (TFC) at 103.87 ± 5.47 mg QE/g, whereas hexane displayed the lowest TPC (16.03 ± 2.22 mg GAE/g) and TFC (8.27 ± 3.88 mg QE/g). The methanolic extract demonstrated the strongest antioxidant activity with an IC50 of 103.3 ± 1.53 μg/mL, compared to the hexane extract's IC50 of 524.7 ± 0.89 μg/mL. Antimicrobial testing revealed that aqueous, ethanolic, and hexane extracts effectively inhibited Klebsiella pneumoniae with a zone of inhibition of 17 mm, comparable to the positive control neomycin (24 mm). The ethanolic extract showed substantial minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against K. pneumoniae and Staphylococcus aureus (3.12 mg/mL and 6.25 mg/mL, respectively), relative to the control (0.0078 mg/mL and 0.015 mg/mL). Additionally, cytotoxicity assays showed LC50 values of 1077.78 μg/mL for the ethanolic and 1905.46 μg/mL for the methanolic extract. These findings suggest that the studied plant extracts, particularly methanolic and ethanolic, have significant bioactive potential, positioning them as promising sources for future drug development against infectious diseases.

Background: Secondary metabolites of plants such as phenol and flavonoids have an extreme potential for neutralizing free radicals. The antioxidant and cytotoxic activities of plants are related to phenolic or flavonoids compounds. The occurrence of drug resistance to antimicrobial drugs has led to use of medicinal herbs in treatment of infections. Antibiotic resistant of Staphylococcus aureus has become a major problem in the treatment of diseases. Objectives: The aim of this study was determination of total phenolic content (TPC) and total flavonoids content (TFC) of Polygonatum orientale Desf and Tilia dasystyla and evaluation of their anti-bacterial effects on Staphylococcus aureus bacterium. Investigation of TPC of P. orientale Desf and T. dasystyla has not been reported before. Methods: Total phenolic and flavonoid content of P. orientale Desf and T. dasystyla extracts were determined using colorimetric methods of Folin-Ciocalteu and aluminum chloride. Antimicrobial activities of the extracts were evaluated by microdilution broth and disc diffusion methods to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. Results: The results showed that TFC of P. orientale Desf with the value of 7.9 ± 0.040 mg/g DW extracted with diluted water solvent and boiling method and TPCs of T. dasystyla with the value of 62.13 ± 0.073 mg/g DW extracted with methanol solvent and boiling method were the highest amount. Methanol extract of P. orientale Desf had more antibacterial activities with the MBC and MIC values of 0.140 mg/mL and 8±0.4 mm zone of inhibition. Conclusion: Tilia dasystyla and P. orientale Desf contain bioactive compounds such as phenolic and flavonoids that can be used as promising option in pharmacognostical studies for treatment of S. aureus infections.

ABSTRACTFicus deltoidea Jack (Moraceae) is a well-known medicinal plant used in customary medication among the Malay people to reduce and mend sicknesses such as ulcers, psoriasis, cytotoxicity, cardioprotective, inflammation, jaundice, vitiligo, hemorrhage, diabetes, convulsion, hepatitis, dysentery injuries, wounds, and stiffness. Ficus deltoidea contains a wide variety of bioactive compounds from different phytochemical groups such as alkaloids, phenols, flavonoids, saponins, sterols, terpenes, carbohydrates, and proteins. The genus Ficus has several hundreds of species, which shows excellent therapeutic effects and a wide variety of helpful properties for human welfare. Searching information was collected by using electronic databases including Web of Science, Science Direct, Springer, SciFinder, PubMed, Scopus, Medline, Embase, and Google Scholar. This review is, therefore, an effort to give a detailed survey of the literature on its pharmacognosy, phytochemistry, phytochemical, and pharmacological properties of Ficus and its important species. This summary could be beneficial for future research aiming to exploit the therapeutic potential of Ficus and its useful medicinal species.

Determination of Phytochemicals of Illicium verum hook and its Antioxidant and Antimicrobial Properties of Ethanol, Methanol, Water and Chloroform Extracts

Illicium verum hook (Star anise), a common ingredient in traditional medicine and most commonly used spice in various cuisines due to its phytochemical content, health benefits such as antioxidant and for its aroma. Rapid increase in interest among natural antioxidant other than synthetic antioxidant lead to various investigations. Therefore a study was conducted to determine phytochemicals of Illicium verum hook, a widely used spice and its antioxidant and antimicrobial properties in 80% ethanol, 80% methanol, water and chloroform extracts. Concentration range of all the crude extracts were analysed in three trials. Among all the extracts ethanol extract showed highest phytochemical content and most of the phytochemicals were absent in chloroform extract. Total Phenolic content and Flavonoid content were determined using Folin–Ciocalteu method and Aluminum chloride assay. Methanol produced highest total Phenolic content and total Flavonoid content among all the extracts, which were 112.587±2.256 mg Gallic acid equivalent (GAE) per gram of dry weight of sample and 211.713±8.679 mg Rutin equivalent (RT) per gram of dry weight of sample. Ethanol extract produced more total Flavonoid content than total Phenol content. Chloroform extract produced least total Phenol content and total Flavonoid content, which were 41.2667±8.495 mg Gallic acid equivalent (GAE) per gram of dry weight of sample and 3.551±1.580 mg Rutin equivalent (RT) per gram of dry weight of sample respectively. Significance difference between total Phenolic content and total Flavonoid content of each extract was determined using regression, statistical analysis. Antioxidant scavenging activity and total antioxidant capacity (TAC) were evaluated using 1-Diphenyl-2-picrylhydrazy (DPPH) radical-scavenging activity,2,2′-azino-di-[3ethyl benzthiazoline sulfate] (ABTS) decolorisation assay, Ferric reducing antioxidant assay (FRAP) and Phosphomolybdate assay (TAC). Methanol extract of Illicium verum hook showed highest antioxidant scavenging activity (51.548 mg/mL) except in FRAP assay. IC 50 values of each assay for methanol extract were, 127.089 mg/mL for DPPH, 51.548 mg/mL for ABTS and 320.476 mg/mL for FRAP. Total antioxidant capacity was high in methanol extract with 39.663 mg Ascorbic acid equivalent per gram of dry weight of sample. Antimicrobial activity of I.verum crude extracts were evaluated against Escherichia coli and Staphylococcus aureus using agar disk diffusion method. Inhibition of bacteria growth by the extract was determined using by the diameter of “zone of inhibition”. Methanol crude extract showed high antimicrobial activity against both S.aureus and E.coli through large zone of inhibition. According to this study methanol was determined as the potential solvent to extract crude oil from I. verum. The efficacy of antimicrobial and antioxidant capacity in I.verum crude extract leads to new source of natural antioxidant, in drug development and processed food industry. Phytochemical Properties and Antibacterial Effects of Salvia multicaulis Vahl., Euphorbia microsciadia Boiss., and Reseda lutea on Staphylococcus aureus and Acinetobacter baumanii

Background: Plantshave long served as a rich source of drugs. Givensomemicroorganisms' acquisition of resistance to the current antibiotics, there is a need for discovering new drugs. Objectives: The aim of the present study was to investigate the phytochemical properties and antibacterial effects of Salvia multicaulis Vahl., Euphorbia microsciadia Boiss., and Reseda lutea against Acinetobacter baumanii and Staphylococcus aureus. Methods: In this experimental study, hydroalcoholic (ethanol 70%) plant extracts were prepared by maceration. Minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs) were determined by CLSI broth microdilution and Muller-Hinton agar assay for each sample, respectively. Total phenolic content was measured by Folin-Ciocalteu colorimetric assay and expressed in terms of gallic acid equivalent and total flavonoid content by aluminum chloride colorimetric method and in terms of rutin equivalent. Results: Findings showed that 1, 4, and 1 mg/mL were derived as MICs and 4, 16, and 8 mg/mL as MBCs for S. multicaulis Vahl., E. microsciadia Boiss., and R. lutea, respectively, against S. aureus; 2, 8, and 2 mg/mL were derived as MICs and 16, 32, and 16 mg/mL as MBCs for S. multicaulis Vahl. R. lutea, and E. microsciadia Boiss., respectively, against A. baumanii. In addition, E. microsciadia Boiss. and S. multicaulis Vahl. were found to contain the highest total phenolic and flavonoid content, respectively. Conclusions: The studied plants that were collected from Chaharmahal and Bakhtiari Province can be used to produce antibiotics due to their phenols and flavonoids and exert antibacterial effects on the studied bacteria. Effect of maturity stage at harvest on antioxidant capacity and total phenolics in kiwifruits (Actinidia spp.) grown in Korea

Six cultivars of kiwifruits grown in Korea, including Actinidia eriantha ‘Bidan’, A. arguta ‘Chiak’, A. arguta ‘Darae No. 2’, A. chinensis ‘Haegeum’, A. chinensis ‘Haehyang’, and A. arguta × A. deliciosa ‘Mansoo’, were harvested at various maturity stages to test whether kiwifruit maturity has an influence on antioxidant capacity or total phenolic and flavonoid contents. Kiwifruit extracts were isolated using absolute methanol and then 80% (v·v-1) aqueous methanol during homogenization. ‘Bidan’, collected at the second harvest stage, contained the greatest amount of total phenolics (775.3 mg gallic acid equivalents·100 g-1 fresh weight) and had the highest antioxidant capacity [816.5, 633.2, and 2,662.7 mg vitamin C equivalents·100 g-1 fresh weight for 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) scavenging, 1,1-diphenyl-2-picrylhydrazyl scavenging, and oxygen radical absorbance capacity assays, respectively] among cultivars tested, while ‘Haehyang’, collected at the first harvest, contained the greatest amount of total flavonoids (13.1 mg catechin equivalents·100 g-1 fresh weight). Kiwifruit cultivar and genotype influenced antioxidant capacity, as well as total phenolic and flavonoid contents. No trend, however, was observed in total phenolic and flavonoid contents, and in the antioxidant capacity with respect to maturity stage. Antioxidant capacity had a higher linear correlation coefficient with total phenolic contents than with total flavonoid contents. The results above suggest that kiwifruits at various maturity stages are a valuable source of phenolics and antioxidants for industrial application and consumer health benefit.

Evaluation of Peronema canescens Leaves Extract: Fourier Transform Infrared Analysis, Total Phenolic and Flavonoid Content, Antioxidant Capacity, and Radical Scavenger Activity

BACKGROUND: Peronema canescens (Sungkai) leaves have been popular in Indonesia which contain various bioactive compounds with empirical therapeutic efficacy in dealing with COVID-19 and various other diseases. Total phenolic and flavonoid content and antioxidant activity using the DPPH method from P. canescens leaf extract have not been studied much. AIM: This research has several objectives. The first is to compare the results of qualitative phytochemical analysis of the ethanol extract of the leaves of P. canescens (EEPL). The second is to measure the total phenol and flavonoid content. The third is to test the FTIR and antioxidant activity of the ethanol extract of P. canescens leaves in vitro using the DPPH method. METHODS: Fresh plant material and simplicia, ethanol extract extracted by maceration method using 96% ethanol as solvent from P. canescens. The Dragendorff’s and Mayer test carried out the qualitative phytochemical analysis, FeCl3 test, Salkowski method, Liebermann–Burchard method, foam test, and NaOH reagent. The total phenolic and flavonoid levels were tested using the Folin–Ciocalteu method. In vitro antioxidant activity was carried out using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. RESULTS: The results of qualitative phytochemical screening showed that alkaloids, flavonoids, saponins, tannins, and steroids were detected in the extract of P. canescens. The spectra from the FTIR test results showed various absorbance peak values indicating the bonding of specific functional groups, namely: 418.12, 599.94, 666.67, 1036.39, 1159.52, 1224.16, 1348.95, 1454.19, 1600.87, 1732.00, 2923.13, and 3353.01 cm-1. In the test results, total phenolic content was as much as 5.64% (mgEAG/g) and total flavonoid content of 142,247 mgEQ/g in a sample of 1 mg extract, which was equivalent to 1 mg quercetin. EEPL has antioxidant activity with the DPPH IC50 method of 116.7865 ppm. CONCLUSION: The data obtained at this time can contribute to the exploitation of P. canescens leaves in the future as one of the nutraceutical products, supplements, and herbal medicines by specific industries related to improving the health status of the world community. The higher the bioactive substance in preparation, the more significant the effect of the pharmacological efficacy response. P. canescens ethanol extract has good total phenolic content, total flavonoid content, and antioxidant action.

Correlation Analysis of Antioxidant Activities with Tannin, Total Flavonoid, and Total Phenolic Contents of Nutmeg (Myristica fragrans Houtt) Fruit Precipitated by Egg white

The nutmeg (Myristica fragrans) flesh extract has a strong antioxidant activity. Therefore, M. fragrans flesh can be developed for functional drinks which are sources rich in antioxidants good for the prevention and treatment of diseases such as cancer and cardiovascular diseases. However, the tannins' content can cause a bitter and sour taste. Therefore, the tannins content should be reduced by the addition of egg white. The purpose of this study is to find out the comparison of antioxidant activity between a combination of M. fragrans flesh extract with various concentrations of egg white and to correlate its antioxidant activities with tannin, total flavonoid, and total phenolic contents. The antioxidant activities were conducted on M. fragrans flesh extract by using DPPH and ABTS radicals. Tannin, total flavonoid, and total phenolic contents from M. fragrans flesh extract were also tested. The M. fragrans flesh extracts without addition egg white have a strong antioxidant in scavenging the stable free radical ABTS (89.980±0.480 µg/mL) and intermediate antioxidant in scavenging the stable free radical DPPH (105.669±0.102 µg/mL). It is followed accordingly by tannin, total flavonoid, and total phenolic contents, namely 14.034±0.100 %w/w TAE, 26.929±0.129 %w/w QE, and 53.164±0.129 %w/w GAE, respectively. Correlation of tannin, total flavonoid, and total phenolic contents, which inhibited DPPH and ABTS radicals had R2 values of about 89.23-97.63%. It showed that antioxidant activity is strongly influenced by the tannin, total flavonoid, and total phenolic contents. Therefore, precipitation from the tannin-protein bond caused antioxidant activities were decreased.

Correlation of Total Phenolic and Flavonoid Contents of Brazilian Honeys with Colour and Antioxidant Capacity

Sixty Brazilian honey samples were analysed for their total phenolic content with the Folin-Denis reagent, total flavonoid content by aluminium chloride method, and antioxidant activity by reaction with 2,2-diphenyl-1-picrylhydrazyl radical. Colour was also classified according to visual analysis and Pfund scale. Linear relationships were observed between colour and flavonoid content, total phenolics and antioxidant capacity, and total flavonoid and phenolic contents. The white-coloured Citrus honey showed the lowest antioxidant activity, while the light ambar Verbenaceae honey showed the highest total phenolics and antioxidant activity. Dark-coloured and polyfloral honeys, though less popularized among consumers, showed average to high antioxidant capacity.

Effect of Different Drying Methods on the Total Phenolic and Flavonoid Content and DPPH Free-Radical Scavenging Activity of Pandanus amaryllifolius Roxb. Planted in Mekong Delta

Aims: This study aimed to investigate the total phenolic and flavonoid content and DPPH free-radical scavenging activity of Vernonia amygdalina planted in Mekong Delta. The optimized conditions for maceration of pandan leaves included drying method, ratio of pandan leaf powder-to-solvent, and extraction time.
 Methodology: The fresh pandan leaves were divided into two equal portions, subjected to different drying methods: shade and oven drying. The dried leaf powder was macerated in ethanol at room temperature. The maceration was conducted with 3 different ratios of pandan leaf powder-to-solvent (w/v) (1:10, 1:15 and 1:20), and the extraction time was 1, 2 and 3 days. The total flavonoid content was determined using aluminum chloride method whereas the total phenolic content was assessed using Folin-Ciocalteu assay. Meanwhile, the antioxidant activity of the plant extracts was quantitatively evaluated using DPPH test.
 Results: The results indicated that the best conditions for maceration of pandan leaves were 1:10 shade-dried leaf powder-to-solvent ratio in 1-day extraction time. Accordingly, the total flavonoid and phenolic content was found to be the highest value of 130.02 ± 2.24 mg QE/g of dried extract and 100.67 ± 1.76 mg GAE/g of dried extract (p < 0.05), respectively. The lowest IC50 of DPPH free-radical scavenging activity of pandan leaf extract was found to be 0.90 ± 0.02 mg/mL (p < 0.05). In addition, the Pearson’s correlation coefficient between IC50 of DPPH free-radical scavenge activity and total flavonoid content was R2 = 0.74 compared to that of phenolic content with the value of R2 = 0.69, indicating that the IC50 of DPPH free-radical scavenge capacity of pandan leaves was influenced chiefly by flavonoid compounds.
 Conclusion: There was a significant difference in phenolic and flavonoid content and DPPH free-radical scavenging activity between shade-dried and oven-dried pandan leaf extracts.

Antiproliferative activity in tumor cell lines, antioxidant capacity and total phenolic, flavonoid and tannin contents of Myrciaria floribunda.

Myrciaria floribunda (H. West ex Willd.) O. Berg, Myrtaceae, is a native plant species of the Atlantic Rain Forest, from north to south of Brazil. The lyophilized ethyl acetate extract from the leaves of M. floribunda was investigated for its antiproliferative activity in tumor cell lines, antioxidant capacity and its total phenolic, flavonoid and tannin contents. Antiproliferative activity was tested in vitro against seven human cancer cells and against immortalized human skin keratinocytes line (HaCat, no cancer cell). Antioxidant activity was determined using 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and oxygen radical absorbing capacity (ORAC) assays and total phenolic, flavonoid and tannin contents were determined by spectrophotometric techniques. Ethyl acetate extract of M. floribunda exhibited antiproliferative activity against cancer cell lines with total growth inhibition (TGI) between 69.70 and 172.10 µg/mL. For HaCat cell, TGI value was 213.60 µg/mL. M. floribunda showed a strong antioxidant potential: EC50 of 45.89±0.42 µg/mL and 0.55±0.05 mmol TE/g for DPPH and ORAC, respectively. Total phenolic content was 0.23±0.013g gallic acid equivalents (GAE)/g extract and exhibited 13.10±1.60% of tannins content. The content of flavonoid was 24.08±0.44% expressed as rutin equivalents. These results provide a direction for further researches about the antitumoral potential of M. floribunda.

Evaluation of total phenol and flavonoid content and antimicrobial and antibiofilm activities of Trollius chinensis Bunge extracts on Streptococcus mutans.

Dental caries is a chronic disease with multiple bacterial infections, Streptococcus mutans is the main cariogenic bacteria. Trollius chinensis Bunge is a common folk medicine in the Xinjiang area of China. In this study, we investigated the total flavonoid content and total phenol content in four types of T. chinensis Bunge extracts and the inhibitory effects of these extracts on S. mutans. Agar diffusion method was used to measure the inhibition zone diameters, and the minimum inhibitory concentration and minimum bactericidal concentration were determined by the twofold dilution method. Water extracts from T. chinensis Bunge and ethanol (30, 60, and 90%) extracts at different concentrations could significantly inhibit the growth of S. mutans. Among them, 30% ethanol extract exhibited the best antibacterial and antibiofilms effect. Biofilm research (crystal violet staining and CLSM) showed that 30% ethanol extract of T. chinensis Bunge plays an important role in inhibiting S. mutans growth and the number of biofilms. The results indicate that T. chinensis Bunge extract has good antibacterial and anti-biofilm activity on S. mutans. It has the potential to be developed for the treatment of caries in clinical application.

Total Phenolic, Flavonoid Contents, Antioxidant and Antimicrobial Activities of Hesperis pendula

Many natural products are used in complementary medicine. Plants are widely used among these natural products. In this study, it was aimed to determine the total phenolic and flavonoid contents, total antioxidant status and antimicrobial activity of Hesperis pendula DC. In this context, the above-ground parts of the plant were extracted with ethanol and methanol. The total antioxidant level of the plant was determined using Rel Assay Diagnostics kits (Megatıp/Türkiye). The total phenolic content was assessed using the Folin-Ciocalteu reagent. Aluminum chloride assay was used to estimate the total flavonoid content. Antimicrobial activity was tested against bacterial and fungal strains by agar dilution method. As a result of the studies, it was observed that the ethanol extract of the plant had higher TAS (Total antioxidant status) (5.707±0.194 mmol/L), TOS (Total oxidant status) (21.646±0.239 µmol/L) and OSI (Oxidative stress index) (0.380±0.017) values. Total phenolic content was higher in ethanol extract (116.78±2.51 mg/g) while total flavonoid content was higher in methanol extract (93.64±2.16 mg/g). It was observed that the ethanol and methanol extracts of the plant inhibited the growth of bacteria at 100-200 µg/mL concentrations. It was determined that ethanol extract inhibited the growth of fungi at 200 µg/mL concentration and methanol extract at 200-400 µg/mL concentrations. In this context, it was determined that H. pendula could be a natural antioxidant and antimicrobial source.

Phytochemical screening, antioxidant and analgesic activities of Croton argyratus ethanolic extracts

Series of experiments were conducted to screen phytochemical constituents, antioxidant and analgesic activities of the ethanolic extracts of the plant Croton argyratus. The extracts obtained from leaves, stem and root of the plant were evaluated for their antioxidant activity by means of 2,2- Diphenyl -1-picrylhydrazyl (DPPH) radical scavenging activity, reducing power and total antioxidant capacity as well as total phenolic and flavonoid contents was studied. To determine analgesic property of the antioxidant rich extract, and formalin induced pain, hot plate and tail flick test were performed. The leaves extract showed the highest value of antioxidant activity based on DPPH radical scavenging activity, reducing power and total antioxidant capacity. The leaf extract also produced the highest total phenolic and total flavonoid content and have a significant activity in late phase of the formalin induced pain test at the dose of 200 mg/kg p.o. However, in the hot plate and tail flick tests, the extract did not show any significant analgesic effects. The results suggested the potential use of C. argyratus plant extracts as a natural source of antioxidant and may act peripherally to relieve pain. BACKGROUND As the longevity of human lives has increased, the geriatric population is increasing demonstrating more number of oral candidal infections due to decreased immunity. Natural products are being investigated to be used to treat oral candidiasis in place of synthetic drugs in selective geriatric cases. Propolis is one such natural product, which is time tested and developed by nature as an antimicrobial agent. Its toxicity is very less. It can be used in multiple forms. It has the antioxidant, anti-inflammatory properties apart from antibiotic properties and potency of the material improves as it matures with time. But the constituents of Propolis differ with the different sources of procurement. We wanted to evaluate the efficacy of the Propolis procured from Hubballi against Candida albicans in this study. METHODS In this invitro study, Hubballi Propolis was extracted by maceration and refluxing. Water and 70 % ethanol were used as extraction solvents. Total Phenolic and flavonoid contents were determined by using Folin - Ciocalteu spectrophotometric method and Aluminium chloride colorimetric method respectively. Antimicrobial sensitivity effect of Propolis was estimated by determining the minimum inhibitory concentration (MIC) of the samples. RESULTS The results of the studies revealed that propolis samples have effective antifungal activity against Candida albicans with MIC range from 0.01mg / ml to 0.03 mg / ml and comparable high total phenolic (TP) and total flavonoid (TF) contents ranging from 175.4 ± 5.7 to 192.2 ± 3.3 and 33.08 ± 10 to 31.73 ± 8.5 mg / ml respectively. CONCLUSIONS Hubballi Propolis can be used for treating Candidal infection. Further, water extract Propolis showed better in total phenolic content (TPC) and total flavonoid content (TFC) than the alcohol extract. This finding is important to overcome the disadvantage of alcohol extract and hence very useful for application in Dentistry, cosmetics and pharmaceutical industry.
