Abstract

OBJECTIVE: Aberrant global DNA methylation in embryos derived from somatic cell nuclear transfer (SCNT) resulted in low efficiency embryonic stem cell generation. Recent studies revealed that miRNAs are capable of inducing genome-wide DNA demethylation of differentiated cells for reprogramming to regain their “stemness”. We test whether somatic cell nuclei demethylated by miRNA and DNA methyltransferase (Dnmt1) knockdown can facilitate efficiency of reprogramming after SCNT.
