Abstract
Objective To investigate the effect of recombinant human erythropoietin (r-HuEPO) on the expressions of intracellular adhesion molecule-1 (ICAM-1), interleukin-6 (IL-6) and IL-10 after traumatic brain injury (TBI) in rats, and explore the anti-inflammatory mechanisms of r-HuEPO. Methods Seventy-five adult male Wistar mrs were randomly divided into sham-operated group (n=5), TBI group (n=35) and r-HuEPO treatment group(n=35), and experimental TBI was induced according to Feeney's method in the latter two groups. Serum levels of IL-6 and IL-10 were determined with enzyme-linked immunosorbent assay and the expression of ICAM-1 protein was detected using iramtmohistochemistry after the injury. Results Compared to the sham-operated group, the expression levels of ICAM-1, IL-6 and IL-10 were significantly increased in TBI group. Application of r-HuEPO significantly reduced the expressions of ICAM-1 and IL-6, but increased the serum level of IL-10 after the injury. Conclusion R-HuEPO may decrease the inflammatory response to brain injury by increasing the expression of IL-10 and inhibiting ICAM-1 and IL-6 expressions to provide neuronal protection after TBI.
