Abstract

Objective To investigate the effect of hyaluronic acid on prostaglandin E2 (PGE2) in rats with cyclophosphamide-incuced cystitis.Methods From June 2012 to December 2012,18 SpragueDawley rats were randomized into control group (normal saline),model group (intraperitoneal injection cyclophosphamide) and intervention group (intravesical 0.8 g/L hyaluronic acid) with 6 rats in each group.Urine PGE2 levels were evaluated by enzyme-linked immunosorbent assay,histological changes were evaluated by hematoxylin and eosin,toluidine blue and immunohistochemistry,and functional changes were evaluated by micturition habits and nociceptive behaviors.Results Compared with the control group,each items in the model group increased significantly (P<0.05).The counts of inflammatory cells and mast cells were 39.50±8.31 and 21.42±3.94 respectively and the number of 24 hours micturition was 32.55±5.74.The levels of PGE2 were (10.53 ± 1.94) pg/ml,and the A of mierosomal prostaglandin E2 synthase-1 (mPGES1) was 26.12±3.51.After treatment of hyaluronic acid,all items decreased significantly (P< 0.05).The counts of inflammatory cells and mast cells were 17.20± 5.13 and 10.11 ± 1.85 respectively.The number of 24 hours micturition was 21.32±3.59.The level of PGE2 was (7.53±1.25) pg/ml,and the A of mPGES1 was 10.59±1.92.Conclusion The inhibition of mPGES1/PGE2 might be one of the ways of hyaluronic acid in treatment of cyclophosphamide-induced cystitis.
