DNA-targeting potential and biomedical applications of selected phenolic Schiff base metal complexes: a molecular biology-oriented supported by computational study

Metal-based compounds continue to attract considerable attention as bioactive agents due to their versatile coordination behavior and ability to modulate biomolecular structure and function. In this study, a phenolic Schiff base ligand, 2-[(5-methoxy-2-hydroxy-phenylimino)methyl]-4-nitrophenol (MHNP), was utilized to synthesize a series of Ru(III), Zn(II), and VO(II) complexes. The structures of the obtained complexes were elucidated using various spectroscopic techniques, and their stability was evaluated to assess suitability for biological applications. Density functional theory (DFT) calculations were performed to provide molecular-level insight into electronic properties and preferred coordination geometries. The combined experimental and theoretical results indicate that the MHNP ligand coordinates through NOO donor atoms, forming octahedral geometry with Ru(III), tetrahedral geometry with Zn(II), and square pyramidal geometry with VO(II) complexes. DNA interaction studies were conducted to evaluate the binding affinity of the complexes toward double-stranded DNA. The results demonstrate enhanced DNA-binding upon metal coordination, with binding modes dependent on the metal center. Notably, the Ru(III) complex exhibited stronger interactions, suggesting groove-binding or intercalative behavior, while Zn(II) and VO(II) complexes showed moderate but stable binding profiles. Structure–interaction relationships highlight the significant role of metal ions in tuning biomolecular recognition. Additionally, the redox properties of Ru(III) and VO(II), along with the redox-modulating nature of Zn(II), suggest potential antioxidant activity through regulation of reactive oxygen species. Overall, these findings indicate that MHNP-based metal complexes are promising multifunctional candidates for antimicrobial, anticancer, and antioxidant applications, offering a rational basis for the development of metal-based therapeutics.

Condensing 2-phenoxyaniline with 5-chlorosalicyldehyde under reflux conditions, a 4-chloro-2-(((2-phenoxyphenyl)imino)methyl)phenol Schiff base has been Synthesized. A zinc complex was synthesized by combining the ligand in a 1:1 molar ratio with zinc sulphateheptahydrate. Mass spectroscopy, NMR, infrared, and elemental analysis were used to characterize the ligand and zinc complex. By measuring the molar conductance, the non-electrolytic character of the complex was confirmed. The zinc ion is coordinated in a pentadentate manner, according to an IR and NMR investigation. Viscosity measurements, absorption and fluorescence spectroscopy were utilized to examine the complex's interaction with CT (calf thymus) DNA. Furthermore, the ligand and complex's ADMET characteristics were ascertained through the use of ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity) study. Calculation of the different electronic parameters of the optimized structure through Density Functional Theory (DFT) indicated the stability of the Zn(II) complex. Molecular docking study reflected the future opportunity for the consideration of Zn(II) complex to fight against Alzheimer and Glaucoma diseases.

End-off compartmental pentadentate Schiff base, 2,6-bis[3′-methyl-2′-carboxamidyliminomethyl(6′,7′)benzindole]-4-methylphenol is synthesized and characterized by 2D NMR experiments and mass spectral techniques. The homodinuclear phenalato bridged end-off compartmental Schiff-base complexes Cu(II), Co(II), Ni(II), Mn(II), Fe(III), VO(IV), Zn(II), Cd(II) and Hg(II) have been prepared by the template method using the precursors 2,6-diformyl-4-methylphenol, 3-methyl(6′,7′)-2-benzindolehydrazide and metal chlorides in 1 : 2 : 2 ratio. The complexes are characterized by IR, NMR, UV-vis, FAB-mass, ESR and TGA techniques. Ni(II), Mn(II) and Fe(III) complexes have octahedral geometry, whereas the Cu(II), Co(II), VO(IV), Zn(II), Cd(II) and Hg(II) complexes have square pyramidal geometry. Low magnetic moment values for Cu(II), Co(II), Ni(II), Mn(II), Fe(III) and VO(IV) complexes indicate antiferromagnetic spin-exchange interaction between two metal centers. The metal complexes have been screened for their antibacterial activity against Escherichia coli and Staphyloccocus aureus and antifungal activity against Aspergillus niger and Fusarium oxysporum.

Synthesis and Antibacterial Activity of Azomethine Ligand and Their Metal Complexes: A Combined Experimental and Theoretical Study

An asymmetrical Schiff base triazole ligand (4-((3-mercapto-5-(naphthalen-1-ylmethyl)-4H-1,2,4-triazol-4-yl)imino)methyl)methoxy) (L) was used to generate novel micro complexes of Cr(III), VO(IV), and Mn(II) ions. Different spectroscopic techniques, including UV-vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), flame atomic absorption, conductivity tests, CHNS elemental analysis, and magnetic susceptibility, were used to determine the structures of the Schiff base micro complexes. The density functional theory (DFT) calculation was screened to consider selected complexes. The observed data indicated their stability, and the expected chemical formula of vanadium(IV) was square pyramidal geometry in VO(L) complex formula. In contrast, the Cr(III) and Mn(II) complexes have octahedral geometry in the formulas. Frontier molecular orbitals calculations (MO) have also been performed to better understand the nature of orbitals, EHOMO, and ELUMO, allowing us to confirm the experimental finding. Pseudomonas aeruginosa and Bacillus subtilis, two types of potentially dangerous bacteria, were subjected to tests to see whether L and its metal complexes have any antibacterial activities or not. All compounds were also tested for their antifungal activity against two different types of fungi, Penicillium spp. and Aspergillus flavus. There is significant action has been noted in all cases for the complexes.

Synthesis, structural, pharmacological and molecular docking simulations studies of Schiff base transition metal complexes procured from acetylacetonyl-4-iminoantipyrine and tyrosine

Schiff base complexes of Cu(II), Ni(II), Co(II), VO(II) and Zn(II) ions have been synthesized by condensation of acetylaceto-4-iminoantipyrine and tyrosine. The structural characterization of compounds has been investigated using elemental analysis, molar conductance and magnetic susceptibility measurements, UV-Vis., FT-IR, ESI-mass, 1H-NMR, 13C-NMR, ESR spectroscopy, cyclic voltammetry, XRD and SEM techniques. The observed analytical data indicate that the metal chelates have the general formula of [ML] type. Powder XRD pattern of Schiff base and its complexes authenticate their crystalline nature. From SEM morphology studies, the reduction grain size of Schiff base in metal chelates was observed which is due to the chelation of the Schiff base. From the IR, electronic absorption spectra and magnetic measurement data, square planar geometry was proposed for the metal complexes except for the [VOL] complex which exhibits square pyramidal geometry. The X-band ESR spectra of [CuL] and [VOL] complexes in DMSO solution were recorded at 77 & 300 K and their spin Hamiltonian parameter values support the proposed geometry. The interactions of the [CuL] complex with CT-DNA were investigated by UV-Vis and absorption titrations. DNA binding interaction studies reveal the hydrophobic interaction between CT-DNA and complexes. Furthermore, analgesic, anti-inflammatory, CNS and antimicrobial studies of the Schiff base and its transition metal complexes were examined and resolved metal complexes have enhanced pharmacological and biological activity when compared to free Schiff base. Molecular docking studies of the complex with DNA and (PDB ID: 6COX) protein were investigated.

A novel metal complex was synthesized using freshly prepared 2-Amino-5-nitro-N-[(E)-thiophen-2-yl-methylidene]aniline ligand with Zn (II) sulphate heptahydrate in a 1:1 molar ratio. The ligand and the complex were characterized using different spectroscopic techniques, and the complex was assigned a distorted square pyramidal geometry. Additionally, DNA binding assays and antibacterial activity were used to assess the biological perspectives for the synthesized complex, including the ligand and complex which was further confirmed by molecular docking. Fluorescence Spectroscopy, viscosity measurement, and adsorption measurement were used to investigate the interaction of the Zn (II) complex with CT-DNA. A comparativein vitroantibacterial activity study againstEscherichia coli, Klebsiella pneumoniae,Bacillus subtilis,andStaphylococcus aureusstrains were studied with free ligand and Zn (II) metal complex. The stable geometry of the complex was additionally established through computational simulation utilizing density functional theory, which was followed by the calculation of several electronic properties. The ADMET characteristics of the complex and ligand were also assessed using ADMET analysis. The in-silico ADMET properties pointed to a significant drug-likeness feature in the synthesized compounds, based on the Lipinski criteria.

A study of DNA/BSA interaction and catalytic potential of oxidovanadium(v) complexes with ONO donor ligands.

The study of DNA/BSA interaction and the catalytic potential of four mononuclear oxidoalkoxido vanadium(v) [VVO(L1-4)OEt] (1-4) and one dinuclear oxidoalkoxido mixed-ligand vanadium(v) [{VO(L2)OEt}2(Q)]{Q = 4,4'-bipyridine}(5) complexes, with tridentate binegative aroylazine ligands are reported [where H2L1 = anthranylhydrazone of 2-hydroxy-1-napthaldehyde, H2L2 = salicylhydrazone of 2-hydroxy-1-napthaldehyde, H2L3 = benzoylhydrazone of 2-hydroxy-1-acetonaphthone, H2L4 = anthranylhydrazone of 2-hydroxy-1-acetonaphthone]. All the complexes are characterized by elemental analysis as well as various spectroscopic techniques. Single crystal X-ray diffraction crystallography of 2 reveals that the metal centre is in distorted square pyramidal geometry with O4N coordination spheres, whereas 5 exhibits a distorted octahedral geometry around the metal center. In addition, all the complexes (1-5) show moderate DNA binding propensity which is investigated using UV-vis absorption titration, circular dichroism, thermal denaturation and fluorescence spectral studies. The experimental results show that the complexes effectively interact with CT-DNA through both minor and major groove binding modes, with binding constants ranging from 104-105 M-1. Among 1-5, complexes 3 and 4 show higher binding affinity towards CT-DNA than others and at the same time also exhibit negative ΔTm values of about ∼1.5 and 1.0 °C which resembles the properties shown by cisplatin. All complexes show moderate photo-induced cleavage of pUC19 supercoiled plasmid DNA with complex 3 showing the highest photo induced DNA cleavage activity of ∼48%. In coherence with the DNA interaction studies, 3 and 4 also exhibit good binding affinity towards BSA in the range of 1010-1011 M-1, which is also supported by their ability to quench the tryptophan fluorescence emission spectra of BSA. All the complexes show remarkable photo-induced BSA cleavage activity (>90%) at a complex concentration of 50 μM. The catalytic potential of 1-5 is also tested for the oxidative bromination of styrene, salicylaldehyde and oxidation of methyl phenyl sulphide. All the reactions show a high percentage of conversion (>90%) with a high turnover frequency (TOF). Particularly, in the oxidative bromination of styrene the percentage of conversion and TOF vary from 96-98% and 8000-19 600 (h-1) respectively, which signifies the potential of these oxidovanadium(v) complexes to stimulate research for the synthesis of a better catalyst.

A new Schiff base ligand obtained from carbazole was synthesized and structurally characterized. Its coordination complexes with Co (II), Ni (II), Cu (II), and Zn (II) ions were prepared for the first time and investigated through various spectroscopic techniques, including ¹H-NMR, UV-Vis, and FT-IR. Density Functional Theory (DFT) estimations were also employed to study the electronic structures, stability, and optimized geometries of the ligand and its complexes. The Co (II) and Ni (II) complexes exhibited an octahedral geometry with a general formula of [M(L)₂Cl₂], while Cu (II) and Zn (II) formed either square planar or tetrahedral geometries, depending on the coordination environment. Analysis of spectral and computational data suggested that nitrogen and oxygen atoms in the ligand actively participated in metal coordination. In antibacterial testing, the synthesized metal complexes demonstrated enhanced inhibition against Klebsiella pneumoniae and Staphylococcus aureus compared to the uncoordinated ligand, suggesting that complexation improved antimicrobial activity. These findings highlight the novelty of the synthesized ligand–metal complexes and their potential in medicinal chemistry, particularly given their distinct structural and electronic characteristics.

A Schiff base ligand HL, (E)-2-((adamantan-1-ylimino)methyl)-6-allylphenol, was synthesized by condensation of amantadine with 3-allyl-2-hydroxybenzaldehyde, followed by the synthesis of its Zn(ii), Co(ii), Cr(iii), and VO(iv) complexes under reflux conditions. The synthesized compounds were comprehensively elucidated by using different spectroscopic and analytical techniques: UV-Vis, 1H and 13C-NMR, FT-IR, ESI-MS, thermal, and single-crystal XRD analysis. The chemical composition of the synthesized compounds was also verified by molar conductance and elemental analysis. An octahedral geometry for Cr(iii) and Co(ii) complexes, tetrahedral for Zn(ii) complex, and square pyramidal geometry have been proposed for VO(iv) complexes. The antidiabetic activities of the synthesized compounds were also evaluated by performing in vitro α-amylase and α-glucosidase inhibition studies. The Co(ii) complex exhibited the highest α-glucosidase inhibitory activity, whereas oxovanadium(iv) and zinc(ii) complexes were also found to be effective against α-amylase. In alkaline phosphatase (ALP) inhibition studies, the HL was found to be inactive, while the complexes showed remarkable enzyme inhibition in the following order: VO > Zn > Co, in a concentration-dependent manner.
