Abstract
An improved direct spectrophotometric method is described that is suited to determinations of sulfide in enzyme mixtures and in biological materials. The assay is based on the specific incorporation of sulfide into methylene blue in the presence of acidic, N,N-dimethyl- p-phenylenedi-amine and ferric chloride. The major improvement in the method is achieved by quantitative precipitation of zinc sulfide and removal of the soluble components of the sample. This procedure replaces the cumbersome entrainment of hydrogen sulfide with nitrogen gas prior to addition of the color-forming reagents. This very simple in situ manipulation is efficient in preventing the interfering effects of foreign constituents of the sample medium and contributes to a further increase in the sensitivity of the method.
