Differential Effects of Aging on Behavior in Working and Untrained Companion Dogs

Aging related declines in cognition and behavior may have a detrimental impact on working dog performance and welfare. However, while aging related changes have been well documented in companion dogs, no work has determined if those findings generalize to working dogs. In the current study, we examined the effects of aging on cognitive and behavioral traits that are important for career success in working dogs using a questionnaire distributed to detection dog handlers/owners. We also included a group of breed-, age-, and sex-matched untrained companion dogs in order to directly compare the effects of age on these traits between working and non-working dogs. The questionnaire comprised a validated measure of impulsivity using the Dog Impulsivity Assessment Scale (DIAS), responses to positive and negative stimuli (Positive and Negative Affect Scale; PANAS), and canine cognitive dysfunction (CCD; Canine Cognitive Dysfunction Rating Scale; CCDR). While detection dogs demonstrated the typical relationship between increased age and prevalence of CCD, it was not associated with retirement, suggesting that detection dogs are not retired due to cognitive decline. Further, in contrast to untrained companion dogs, detection dogs demonstrated no aging related declines in aspects of behavior, including those shown to be important for working success (e.g., energy and interest). These findings are the first to demonstrate a differential effect of aging on some aspects of behavior between working and non-working dogs. We discuss these results in relation to previous findings on the declines in similar traits shown in companion dogs.

Enhanced Selection of Assistance and Explosive Detection Dogs Using Cognitive Measures

Working dogs play a variety of important roles, ranging from assisting individuals with disabilities, to explosive and medical detection work. Despite widespread demand, only a subset of dogs bred and trained for these roles ultimately succeed, creating a need for objective measures that can predict working dog aptitude. Most previous research has focused on temperamental characteristics of successful dogs. However, working dogs also face diverse cognitive challenges both in training, and throughout their working lives. We conducted a series of studies investigating the relationships between individual differences in dog cognition, and success as an assistance or detection dog. Assistance dogs (N = 164) and detection dogs (N = 222) were tested in the Dog Cognition Test Battery, a 25-item instrument probing diverse aspects of dog cognition. Through exploratory analyses we identified a subset of tasks associated with success in each training program, and developed shorter test batteries including only these measures. We then used predictive modeling in a prospective study with an independent sample of assistance dogs (N = 180), and conducted a replication study with an independent sample of detection dogs (N = 90). In assistance dogs, models using data on individual differences in cognition predicted higher probabilities of success for dogs that ultimately succeeded in the program, than for those who did not. For the subset of dogs with predicted probabilities of success in the 4th quartile (highest predicted probability of success), model predictions were 86% accurate, on average. In both the exploratory and prospective studies, successful dogs were more likely to engage in eye contact with a human experimenter when faced with an unsolvable task, or when a joint social activity was disrupted. In detection dogs, we replicated our exploratory findings that the most successful dogs scored higher on measures of sensitivity to human communicative intentions, and two measures of short term memory. These findings suggest that that (1) individual differences in cognition contribute to variance in working dog success, and (2) that objective measures of dog cognition can be used to improve the processes through which working dogs are evaluated and selected.

The assisted reproductive techniques used in dogs have strictly limited utility when compared with other mammals. Although successful somatic cell cloning has been reported, artificial insemination by frozen semen has been only readily available method for improved breeding for companion and working dogs. Recently, successful cryopreservation of embryos and subsequent embryo transfer with a non-surgical technique in dog was reported. Application of embryo cryopreservation and transfer technology could contribute to breeding management in companion dogs, working dogs including guide dogs and drug-detecting dogs and quarantine dogs. Such technology would also facilitate the transportation and storage of genetic materials and aid in the elimination of vertically transmitting diseases in the dog.

Enhancing the Selection and Performance of Working Dogs

Dogs perform a variety of integral roles in our society, engaging in work ranging from assistance (e.g., service dogs, guide dogs) and therapy to detection (e.g., search-and-rescue dogs, explosive detection dogs) and protection (e.g., military and law enforcement dogs). However, success in these roles, which requires dogs to meet challenging behavioral criteria and to undergo extensive training, is far from guaranteed. Therefore, enhancing the selection process is critical for the effectiveness and efficiency of working dog programs and has the potential to optimize how resources are invested in these programs, increase the number of available working dogs, and improve working dog welfare. In this paper, we review two main approaches for achieving this goal: (1) developing selection tests and criteria that can efficiently and effectively identify ideal candidates from the overall pool of candidate dogs, and (2) developing approaches to enhance performance, both at the individual and population level, via improvements in rearing, training, and breeding. We summarize key findings from the empirical literature regarding best practices for assessing, selecting, and improving working dogs, and conclude with future steps and recommendations for working dog organizations, breeders, trainers, and researchers.

Musculoskeletal injuries can lead to a working dog being withdrawn from service prior to retirement. During training exercises, young working dogs are often required to perform repetitive tasks, including adoption of an upright posture (or “hupp” task). Non-invasive, quantitative methods would be helpful for supporting research on effects of these repetitive tasks on sacroiliac joints (SIJ). Furthering our understanding of lesions in and biomechanical stresses on the SIJ could provide insight into possible training modifications for minimizing risks of SIJ injury. Aims of this retrospective, secondary analysis, exploratory study were to test hypotheses that (1) mean numbers of SIJ computed tomographic (CT) lesions/dog would differ among work status groups in young working Labrador Retrievers; (2) a methodology for using CT data and finite element analysis (FEA) to quantify SIJ ligament strain in the static canine pelvis would be feasible; and (3) this FEA methodology would yield repeatable measures of SIJ ligament strain. Clinical and CT data for 22 Labrador retriever working dogs, aged 11–48 months, were retrospectively reviewed. Dogs were categorized into three work status groups (Breeder, Detection, Other). A veterinary radiologist who was unaware of dog group status recorded numbers of CT lesions for each SIJ, based on previously published criteria. Mean numbers of SIJ CT lesions/dog were compared among dog work status groups. An a priori FEA model was created from the CT images of one of the dogs using image analysis software packages. Using tissue properties previously published for the human pelvis, various directional loads (n = 8) and forces (48 ligament strain values) were placed on the canine model in five trials. Repeatability was tested using regression analysis. There was a significantly greater mean number of subchondral sclerosis lesions in left SIJ of Breeder vs. Detection dogs, a significantly greater mean number of subchondral cysts in right SIJ for Detection vs. Breeder dogs, and a significantly greater mean number of subchondral cysts in right SIJ of Other vs. Breeder dogs (p < 0.05). Finite element modeling and analysis using CT data was feasible and yielded repeatable results in 47/48 (98%) of tests at each combination of strain, ligament, and side.

Simple SummaryEarly attachments to human carers may have long lasting impacts on a seeing eye dog’s working life. Using a self-report questionnaire, we found that puppy carers demonstrated more avoidant styles of attachment towards the dogs in their care as compared to dog owners. Carers also predominately appeared to be driven to the puppy caring role by more pro-social motivations that do not relate to the dog itself. Carers’ reluctance to form close relationships with the puppies in their care may impact subsequent bond formation with other humans, as well as their working performance and wellbeing.Important physiological, performance, and relationship differences have been reported between companion and working dogs. This study aimed to investigate how human attachment styles manifest towards dogs, depending on the dog’s role. Seeing Eye Dog (SED) carer (n = 25) and Companion Dog Owner (CDO) (n = 78) avoidant and anxious attachment scores towards the dog in their care were compared. Feelings and motivations about being a SED carer or CDO were also investigated qualitatively. Significant differences were observed between pet avoidance, with avoidance scores significantly predicting SED carer group membership. Qualitative insights revealed more human prosocial motivations for becoming a SED carer, and more dog-related motivations for becoming a CDO, with CDOs more likely to consider their dog a ‘significant other’. This study corroborates findings supporting differences in human–dog relationships for working versus companion dogs. The potential impacts of human–dog attachment should be considered for SED success.

An examination of attitudes and kennel management practices relating to the welfare of working dogs.

This thesis explores how people value the welfare of dogs and what they think is important to the welfare of dogs living in kennels. This research shows that young working dogs find training kennel facilities to be stressful. Access to an enrichment program did not increase stress and may help young working dogs to complete their training. The research also identified better ways to measure stress in dog welfare research using salivary cortisol. The thesis proposes that assuring the public that working dogs have good welfare will be important to the sustainability of relevant industries.

The assisted reproductive techniques (ARTs) such as in vitro fertilization, embryo transfer, and cryopreservation of gametes have contributed considerably to the development of biomedical sciences in addition to improving infertility treatments in humans as well as the breeding of domestic animals. However, ARTs used in canine species have strictly limited utility when compared with other mammalian species, including humans. Although successful somatic cell cloning has been reported, artificial insemination by frozen semen to date is only available for the improved breeding and reproduction for companion and working dogs as well as guide dogs for the blind. We describe here the successful cryopreservation of embryos and subsequent embryo transfer in dogs. Canine embryos were collected from excised reproductive organs after artificial insemination and subsequently cryopreserved by a vitrification method. When the 4-cell to morula stage of cryopreserved embryos were nonsurgically transferred into the uteri of nine recipient bitches using a cystoscope, five recipients became pregnant and four of them delivered a total of seven pups. The cryopreservation of embryos in canine species will facilitate the transportation and storage of genetic materials and will aid in the elimination of vertically transmitted diseases in dogs. In addition, this technique will contribute to the improved breeding of companion and working dogs such as guide dogs, drug-detecting dogs, and quarantine dogs.

Energy intake and expenditure of improvised explosive device detection dogs

Improvised explosive device detection (IDD) dogs explore up to 40 km of land daily and therefore have energetic demands that may be above the National Research Council’s requirement for working dogs. This study was designed to quantify metabolic energy intake (MEI) and total energy expenditure (TEE) in a group of IDD dogs. Two groups of dogs that had undergone different training protocols (CP1, n=8 and CP2, n=11) underwent a 5-day deployment simulation that consisted of combined road clearing, orbit and point-to-point activities and lasted approximately 9 h per day. The CP1 dogs were fed according to the IDD Marine Corps Manual, while CP2 dogs were offered additional calories based on pilot study data of energy expenditure. The MEI was calculated based on feed intake rates and chemical composition of the diets. TEE was quantified using the doubly-labelled water technique in 2 of the CP1 dogs and 7 of the CP2 dogs. During the 5-day deployment simulation the MEI ranged from 189-310 kcal/bodyweight (BW)0.75 per day, with the CP2 dogs at the higher end because they were offered more feed. The TEE ranged between 375-507 kcal/BW0.75 per day, above the MEI, suggesting the dogs were in negative energy balance and metabolic reserves within the body were combusted for energy production. These findings reveal that energy requirements of deployed military working dogs are higher than previously published metabolic energy requirements of working dogs.

Canine Cognitive Dysfunction Is Associated with Alzheimer’s Disease-Related Changes in the Brain Transcriptome and RNA Changes in Plasma Extracellular Vesicles

Chronic, age-related diseases, such as Alzheimer’s disease (AD), can be diffcult to study longitudinally because humans have long lifespans and are highly heterogeneous. Recently, companion dogs have emerged as a promising model of AD because dogs: a) have a shorter lifespan than humans; b) are heterogeneous like humans; c) exposed to diverse human environments; and d) naturally develop canine cognitive dysfunction (CCD), similar to AD. However, the transcriptomic similarities between AD and CCD have not been evaluated, and emerging mechanisms of AD have not been explored in dogs. One such mechanism involves extracellular vesicles (EVs), ~100nm lipid bilayer particles that can package neurodegeneration-relevant cargo (e.g., RNAs, cytokines), export it to other cells, and contribute to aging/AD-relevant processes like inflammation. To better understand CCD and determine if EVs may play a role in it, we generated total transcriptome (RNA-seq) on cortex tissue and EVs isolated from plasma of young, old, and CCD dogs, and we evaluated cytokine expression in corresponding plasma samples via ELISAs. In the animals we studied, we observed functional differences between old healthy and CCD animals, reflected by Canine Dementia Scale scores (CADES; 6 and 57, respectively). In our RNA-seq analyses, we found marked age- and CCD-related transcriptome differences in the cortex (328 and 2673 differentially expressed genes, respectively), which mostly reflected changes in immune activation and neuronal health (similar to what has been observed in AD). Moreover, when we overlapped transcriptome data on the cortex and plasma EVs, we found a significant overlap in genes that were either up- or down-regulated in both samples (344 and 515, respectively). The processes related to these genes reflected changes in regulation of immune responses, as well as synaptic signaling. We also found evidence of CCL5 (RANTES, increased in AD) RNA in plasma EVs only in CCD samples, which corresponded with increased CCL5 expression in the brains of the same animals, although no differences were observed in plasma concentrations. While preliminary in nature, these results suggest that CCD is associated with AD-like transcriptomic changes in the brain, and with AD-relevant changes in EV cargo. Future studies are needed to determine the functional relevance of these data, but our results suggest the possibility of finding specific EV/RNA markers that could predict disease severity in CCD, and perhaps eventually in AD.
