Abstract
An analytical method was developed for bisphenol A (BPA) in canned fish and meat. A sample was extracted with 150mL of ethyl acetate and the organic layer was washed with 150mL of 5% NaCl solution. After defatting by n-hexane/acetonitrile partition, the extract was loaded on a 10% water-dehydrated silica gel column, washed with 100mL of 15% acetone/n-hexane and eluted with 50mL of 60% acetone/n-hexane. The eluate was evaporated to dryness and dissolved in methanol. BPA was analyzed by HPLC [column, Wakosil 5C18 (4.6mm i. d.×150mm); mobile phase, 60% methanol; detection, 230nm; flow rate, 0.8mL/min].Recovery of BPA in 20g of canned tuna steak (maguro steak) fortified with 1μg of BPA was 89.9±7.1 (n=3). Four kinds of canned fish and three kinds of canned meat on the market were analyzed for BPA with the developed method. The ranges of BPA content were N. D. -319ng/g. The detection limit of BPA was 25ng/g.This method was effective for BPA in canned fish and meat and does not use solvents such as chloroform and dichloromethane.
