Abstract

Isolated rat lungs were perfused «in vitro in a Delaunois apparatus with 14-14C erucate and 9–10. 3H oleate, during 30 or 60 min. The perfusate was collected at the output of lungs and not re-used. The 3H and 14C radioactivities were determined in the perfusate and lung lipids and in phospholipid and neutral lipid fatty acids of these fractions. Oxydative cleavage of the double bond of the labelled monounsaturated fatty acids of phospholipids was also performed and 14C radioactivity of the residual compounds determined. At the end of experiments loss of 14C radioactivity was greater than that of 3H. More 14C radioactivity was recovered in lung lipids. The free fatty acid fraction was generally more labelled and it contained more 14C than 3H. Phospholipids and triglycerides were labelled to about the same extent by both 14C and 3H. The 3H radioactivity was mainly recovered in oleic acid; next the most labelled fatty acids were palmitoleic acid and 20:1 but the conversion rate was very low. On the contrary, 14C erucic acid was more extensively converted into monounsaturated fatty acids, mainly in the phospholipid fraction. It is mainly converted in decreasing order into 18:1 Δ9 (oleic acid), 16:1 Δ7 (an isomer of palmitoleic acid), 24:1 Δ15 (nervonic acid) and 20:1 Δ11 (an isomer of gadoleic acid). These fatty acids are derived from erucic acid by chain lengthening or shortening from its carboxylic end. On perfuse des poumons de rats isolés maintenus en survie dans l'appareil de Delaunois, par de l'érucate 14 14C1 et de l'oléate 9–10 3H2. La perfusion ouverte dure 30 ou 60 mn. On mesure la radioactivité 3H et 14C des lipides du perfusat, du poumon, des fractions lipidiques pulmonaires ainsi que celle des acides gras des phospholipides et des lipides non phosphorés. On détermine en outre la radioactivité 14C présente sur les fragments provenant de la coupure oxydative de la double liaison des acides monoinsaturés des phospholipides. La perte de radioactivité 14C en fin de perfusion est supérieure à la perte de radioactivité 3H. On retrouve toujours plus de 14C que de 3H dans les lipides pulmonaires. La fraction acides gras libres est en général la plus marquée et renferme toujours plus de 14C que de 3H. Les autres fractions les plus marquées sont ensuite les triglycérides et les phospholipides. La radioactivité 3H se retrouve essentiellement sur l'acide oléique, les acides les plus marqués étant ensuite le 16:1 et le 20:1. L'acide érucique est par contre converti de façon plus importante en acides gras monoinsaturés. Il est principalement transformé, par ordre d'importance: en 18:1Δ9 (oléique), 16:1Δ7, 24.1Δ15 (nervonique) et 20:1Δ11. Ces acides gras proviennent de l'élongation ou du raccourcissement de la chaîne de l'acide érucique à partir de son groupement carboxylique.
