Anti-Klebsiella and antibiotic-potentiation activities of the methanol extracts of seven Cameroonian dietary plants against multidrug-resistant phenotypes over-expressing AcrAB-TolC efflux pumps

Background: Bacterial infections continue to wreak havoc around the world with high death rates. African medicinal plants and the phytoconstituents showed high efficiency in impeding the growth of the resistance phenotypes of bacteria The present work was designed to determine the antibacterial activity of seven Cameroonian dietary plants against clinical MDR isolated of Klebsiella sp. These plants included Persea americana Miller (Lauraceae), Psidium guajava Linn. (Myrtaceae), Mangifera indica Linn. (Anacardiaceae), Citrus sinensis Linn. (Rutaceae), Passiflora edulis Sims (Passifloraceae), Garcinia kola Heckel (Guttiferae), and Artocarpus heterophylus Lam. (Moraceae). Methods: Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) determinations on the used bacterial strains alone, in the presence of an efflux pump inhibitor, phenylalanine arginine beta naphthylamide (PAβN), and in combination with antibiotics, were performed by the microbroth dilution method using a rapid colorimetric para-Iodonitrotetrazolium chloride (INT) assay. Results: The tested botanicals had different extend of antibacterial activities, with MIC ranges of between 256 μg/mL and 2048 μg/mL. Botanicals from Persea americana, Psidium guajava, Mangifera indica, Artocarpus heterophyllus, and Garcinia kola bark had detected MIC values on all 15 tested Klebsiella strains. PAβN potentiated the activity of the botanicals on all tested bacteria, with the increase of activity ranging from 4 to more than 128-fold. The most significant increase of 4 to more than 128-fold was observed with botanicals from leaves and bark of Psidium guajava and Mangifera indica. The botanicals from the leaves of Mangifera indica potentiated the activity of eight out of ten tested antibiotics (Ceftriaxone, Chloramphenicol, Levofloxacin, Ampicillin, Tetracycline, Imipenem, Doxycycline, and Levofloxacin) against 100% of the tested bacteria. Conclusion: In the present study, it was demonstrated that botanicals from Persea Americana, Psidium guajava, Mangifera indica, Artocarpus heterophyllus, and Garcinia kola had the highest spectrum of activity, and can be used to combat the resistance of Klebsiella species

Effects of Incorporating Mango (<i>Mangifera indica</i>), Avocado (<i>Persea</i><i> americana</i>) and Guava (<i>Psidium guajava</i>) Leaves Powder in the Ration on Growth Characteristics in Rabbits

A study conducted on burgundy rabbits evaluated the effects of rations containing mango (Mangifera indica), avocado (Persea americana) and guava (Psidium guajava) powder. For this purpose, 36 rabbits (20 males and 16 females) aged 60 to 70 days with an average weight of 1012 ± 133 g were divided randomly into 4 equal groups (T0, T1, T2 and T3) of 9 animals each according to body weight (bw). The rabbits were divided throughout the trial, the animals were fed, those of group T0 (control) with a standard feed without any type of leaf powder, while those of groups 1, 2 and 3 received the standard feed supplemented with Mangifera indica, Persea americana and Psidium guajava leaves powder at the rate of 0.5% respectively. After two months of treatment, the animals were fasted for 12 hours (20 h - 8 h) and sacrificed to evaluate carcass and digestive organ weights. The results revealed that the values of feed intake, live weight and weight gain were non-significantly (p > 0.05) increased in all animals regardless of the ration in reference to the control. The different leaves powder induced an increase in carcass yield in rabbits but this increase was only significant (p M. indica leaf powder (51.03 ± 0.24) as compared to the control (48.63 ± 0.74). Gut length was significantly increased in animals fed rations containing Psidium guajava and Persea americana leaves powder compared to the control. A significant increase in gut weight was observed in animals fed the Persea americana leaf supplemented ration. In general, mango leaf powder performed better than other types of powders.

Mangifera indica 'Namdokmai' Prevents Neuronal Cells from Amyloid Peptide Toxicity and Inhibits BACE-1 Activities in a Drosophila Model of Alzheimer's Amyloidosis.

Alzheimer’s disease (AD) is a progressive neurological illness with few effective treatments. Thus, ameliorating the effects of AD using natural products has attracted global attention with promising efficacy and safety. In this study, ten tropical fruits including Ananas comosus ‘Phulae’, Ananas comosus ‘Pattavia’, Carica papaya ‘Khaekdum’, Carica papaya ‘Khaeknuan’, Durio zibethinus ‘Monthong’, Durio zibethinus ‘Chanee’, Psidium guajava ‘Kimju’, Psidium guajava ‘Keenok’, Mangifera indica ‘Kaew’ and Mangifera indica ‘Namdokmai’ were screened for their inhibitory activities against the key enzymes, cholinesterases and β-secretase (BACE-1), involved in AD pathogenesis. The top three fruit extracts with promising in vitro anti-AD activities were further investigated using rat pheochromocytoma PC-12 neuronal cell line and Drosophila AD model. Data showed that M. indica ‘Kaew’, M. indica ‘Namdokmai’ and P. guajava ‘Kimju’ reduced Aβ1–42-mediated neurotoxicity by promoting glutathione-dependent enzymes, while M. indica ‘Namdokmai’ limited Aβ1–42 peptide formation via BACE-1 inhibition and amended locomotory behavior of the Drosophila AD model. Results indicated the potential anti-AD properties of tropical fruits, particularly M. indica ‘Namdokmai’ in the prevention of Aβ1–42-mediated neurotoxicity and as a BACE-1 blocker.

Antibacterial activity of extracts of twelvecommon medicinal plants from the Philippines

The antibacterial activity of the n-hexane, acetone/dichloromethane, ethanol and aqueous extracts of twelve common medicinal plants from the Philippines obtained through pounding and solvent extraction was evaluated using disc Agar diffusion. The microorganisms tested were: Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa. Susceptibility test results showed that different extracts (1000 mcg/disc) from ten plants inhibited growth with a mean zone of inhibition range of 15 to 30 mm against S. aureus, E. coii, and P. aeruginosa. The common medicinal plants which showed highly positive activity were Psidium guajava (guava), Eucalyptus globulus, Mangifera indica(Indian mango), Nasturtium officinale (Watercress), Pterygospermum oleiferum (Moringa),Carmona retusa (Wild tea), Citrus aurantifolia (Lemon), Citrus sinensis (Orange), Allium sativum (garlic), and Allium cepa (onion). Preliminary phytochemical screening revealed the presence of flavonoids, tannins, alkaloids, glucosides, saponins and steroids/terpenes. The results suggest that the different plant extracts contain bioactive constituent(s) particularly tannins, flavonoids, terpenoids and other glycosides with very strong antibacterial activity and validates the ethno-medical use in the treatment of bacterial skin diseases and other forms of bacterial infections. However, many reputed plant antibacterials have very variable activities depending on certain conditions. A Review of Air Pollution Mitigation Approach Using Air Pollution Tolerance Index (APTI) and Anticipated Performance Index (API)

Air pollution is a global environmental issue, and there is an urgent need for sustainable remediation techniques. Thus, phytoremediation has become a popular approach to air pollution remediation. This paper reviewed 28 eco-friendly indigenous plants based on both the air pollution tolerance index (APTI) and anticipated performance index (API), using tolerance level and performance indices to evaluate the potential of most indigenous plant species for air pollution control. The estimated APTI ranged from 4.79 (Syzygium malaccense) to 31.75 (Psidium guajava) among the studied indigenous plants. One of the selected plants is tolerant, and seven (7) are intermediate to air pollution with their APTI in the following order: Psidium guajava (31.75) > Swietenia mahogany (28.08) > Mangifera indica L. (27.97) > Ficus infectoria L. (23.93) > Ficus religiosa L. (21.62) > Zizyphus Oenoplia Mill (20.06) > Azadirachta indica A. Juss. (19.01) > Ficus benghalensis L. (18.65). Additionally, the API value indicated that Mangifera indica L. ranges from best to good performer; Ficus religiosa L. and Azadirachta indica A. Juss. from excellent to moderate performers; and Cassia fistula L. from poor to very poor performer for air pollution remediation. The Pearson correlation shows that there is a positive correlation between API and APTI (R2 = 0.63), and this implies that an increase in APTI increases the API and vice versa. This paper shows that Mangifera indica L., Ficus religiosa L., and Azadirachta indica A. Juss. have good potential for sustainable reduction in air pollution for long-term management and green ecomanagement development.

Urban air pollution control: selection of trees for ecological monitoring using anticipated performance indices in a medium-size urban area in Southwest Nigeria

The study assessed the biochemical parameters, sensitivity and tolerance levels to air pollution of the leaves of selected tree species along major roads in Ile-Ife in order to determine the tree species that will be most suitable in developing greenbelts along roadsides. The air pollution tolerance index (APTI) ranged from 8.5 to 13.9 for the entire tree species studied with Gmelina arborea, Polyalthia longifolia, Mangifera indica, Terminalia catappa and Tectona grandis having the highest during the wet season, and Polyalthia longifolia with the APTI in the dry season. Newbouldia laevis, Spondias mombin, Gliricidia sepium, Senna siamea, Psidium guajava and Anacardium occidentale, Albizia zygia were classified as tree species sensitive to air pollution during both dry and wet seasons. With reference to the API grading, Mangifera indica, Tectona grandis, Gmelina arborea with percentage grading of 62.5% were classified as 'good' greenbelt performers. Anacardium occidentale, Terminalia catappa, Spondias mombin, Newbouldia laevis, Senna siamea, Polyalthia longifolia with percentage grading of 56.25%, each, were classified as 'moderate' performers while Gliricidia sepium (50%), Psidium guajava (50%) and Albizia zygia (43.75%) were classified as 'poor' performers for bio-indicators of air pollution. The study concluded that Gmelina arborea, Tectona grandis and Mangifera indica were good biomonitors for greenbelt development.

Comparative evaluation of antibacterial efficacy of plants traditionally used as antidiarrheal against enteropathogenic Escherichia coli

The study was conducted to investigate the antibacterial efficacy against enteropathogenic E. coli of plants commonly used to treat calf-diarrhoea. Methanolic extracts of six plants (Aegle marmelos, Curcuma longa, Dalbergia sissoo, Mangifera indica, Psidium guajava and Punica granatum) were screened for their antibacterial property against enteropathogenic E. coli by standard disc diffusion method. Minimum inhibitory concentration (MIC) and of the extract exhibiting highest antibacterial activity was estimated by broth dilution method and minimum bactericidal concentration (MBC) was measured by streaking the contents of MIC tubes on nutrient agar plates. Among the six extracts tested, only extracts of Curcuma longa, Psidium guajava and Punica granatum exhibited antibacterial activity against E. coli. Out of these three, Punica granatum extract was found to be most effective with a mean inhibition zone of 14.67±0.577 mm followed by Psidium guajava (9.67±0.577 mm) and Curcuma longa (8.67±0.577 mm), produced by the disc containing 8.00 mg of respective extract. Minimum inhibitory concentration and minimum bactericidal concentration of the Punica granatum extract were estimated to be 02.00 mg/mL and 03.00 mg/mL respectively. These findings suggest that methanolic extracts of Curcuma longa, Psidium guajava and Punica granatum possess antibacterial activity against enteropathogenic E.coli

Bioactive Fractions Isolated from By-Products of the Guava (Psidium guajava) and Mango (Mangifera indica L.) Agri-Food Industry

Valorizing agri-food industrial waste is essential for a circular economy, yielding high-value products, waste reduction, technological solutions, employment opportunities, and enhanced food security. This work shows the valorization of seeds generated as residues from the agri-food industries of guava pera (Psidium guajava) and Tommy Atkins mango (Mangifera indica L.), through extraction with supercritical carbon dioxide (scCO2). After the optimization of the initial solid condition of the raw material (i.e., particle size and moisture content), scCO2 pressure and temperature were established through the response surface methodology (RSM) to obtain an oily extract with the highest content in bioactive compounds of commercial relevance, as well as with a high antioxidant capacity. The total amount of oily extract in guava and mango seeds was 14% and 9%, respectively, while the maximum recovery of supercritical extract was 95% from guava seeds at 38 MPa and 50 °C, and 88% from mango seeds at 37 MPa and 63 °C. Bioactive fractions rich in squalene, γ-tocopherol, α-tocopherol, campesterol, β-sitosterol, and stigmasterol were obtained. The best supercritical extraction conditions, in terms of the bioactive fractions richest in minor compounds, were at 17 MPa and 50 °C for guava seeds and at 23 MPa and 63 °C for mango seeds. At these conditions, the highest antioxidant capacities were also found for the extracts. Thus, these bioactive fractions could be used in a variety of products in the cosmetic, food, pharmaceutical, and medical activities due to the beneficial properties of the identified compounds in health as antioxidants, anti-inflammatories, and cholesterol reducers.

GC–MS, antioxidant, anti-inflammatory and in silico studies of the polyherbal formulation containing Mangifera indica, Psidium guajava, Cymbopogun citratus and Azadirachta indica

BackgroundChronic oxidative stress is linked to diseases such as cardiovascular conditions, diabetes, and neurological disorders. Chronic inflammation is related to disorders such as arthritis, asthma, and inflammatory bowel disease. Increasing research indicates that polyherbal formulations effectively mitigate oxidative stress and inflammation. This work focuses on the GC–MS, antioxidant, anti-inflammatory, molecular docking, ADMET (Absorption, Distribution, Metabolism, Excretion, and Toxicity), and molecular dynamics analyses of the polyherbal formulation (PHF), derived from Mangifera indica, Psidium guajava, Cymbopogon citratus, and Azadirachta indica (MPCA).ResultsGC–MS results indicated that the principal compounds include capsaicin (CAP) (5.85%), 5,5-diethylspiro [2.3]hexan-4-one (SHD) (5.40%), chlorogenic acid (CLA) (5.42%), and cyclohexanone, 2-(2-propenyl)- (CLP) (5.70%). MPCA-PHF has higher antioxidant activity in DPPH assays (IC50, 22.47 ± 0.16 µg/mL) but lower when compared to ascorbic acid in the FRAP assay. Indomethacin had better activity compared to MPCA-PHF in terms of membrane stabilisation, but MPCA-PHF is more efficacious in the heat-induced haemolysis assay (IC50, 377.98 ± 2.66 μg/mL) compared to indomethacin (IC50, 549.97 ± 0.60 µg/mL). CAP and CLA had the highest binding affinities of − 8.4 and − 8.6 kcal/mol, signifying robust interactions with the target proteins 2X08 and 1CX2. The MD simulation demonstrated that the hit compound exhibited a stable and flexible structure with the 2XO8 protein. Furthermore, the MPCA PHF compounds exhibited favourable ADMET predictions.ConclusionsThe polyherbal formulation demonstrated significant antioxidant and anti-inflammatory activity, corroborated by molecular docking and dynamics experiments indicating favourable ADMET characteristics, thereby positioning it as a viable candidate for preclinical and clinical research.

Polyisoprenoid Compounds from Tropical Fruit Trees in Universitas Sumatera Utara Campus

Tropical trees are a source of secondary metabolite compounds that have various biological activities that can help human life both for industrial and pharmaceutical needs. One of the secondary metabolites is polyisoprenoid. This study aims to identify and analyze polyisoprenoid compounds quantitatively from tropical fruit trees at the Universitas Sumatera Utara. Nine samples of tropical fruit were obtained, namely: Psidium guajava, Tamarindus indica, Manilkara kauki, Morinda citrifolia, Mangifera indica, Artocarpus communis, Artocarpus heterophyllus, Gmelina arborea, and Syzygium aqueum. Each sample was extracted and isolated the polyisoprenoid alcohol, then analyzed using two-dimensional thin-layer chromatography. The polyisoprenoid in fruit samples found at the Universitas Sumatera Utara was a type II and III polyisoprenoid compound. The total lipid values ranged from 48.7 mg/g dw to 262.9 mg/g dw, polyisoprenoids ranged from 1.2 mg/g dw to 9.5 mg/g dw, and polyprenols ranged from 0.5 mg/g dw to 5.7 mg/g dw. Carbon chain length and dolichol polyprenol lengths of each fruit were collected (Psidium guajava L.) (C70 C80), (Tamarindus indica L.) (C80 C95), (Manilkara kauki L.) (C80 C95 and C70 C90), (Morinda citrifolia L.) (C75 C100), (Mangifera indica L.) (C80 C90 and C75 C90), (Artocarpus communis) (C75 C95), (Artocarpus heterophyllus) (C70 C75 and C70 C100), (Gmelina arborea Roxb.) (C90 C95), and (Syzygium aqueum) (C65 C90 and C75 C90).

Heavy Metal Resistance by Endophytic Bacteria Isolated from Guava (Psidium Guajava) and Mango (Mangifera Indica) Leaves

Heavy metal resistant bacteria are widespread in nature and their application in decontamination of polluted ecosystems is promising. In this study, ability of endophytic bacteria isolated from Psidium guajava (Guava) and Mangifera indica (Mango) for heavy metal resistance was assessed. Leaves samples form the two plants were collected and processed according to the standard laboratory practices. Heavy metals were analyzed using Atomic absorption spectrophotometer. Endophytic bacteria were isolated and identified using morphological and biochemical characteristics; heavy metal resistance was determined by plate dilution method. Heavy metal analysis revealed that the leaves samples contained considerable quantities of Manganese (Mn), Lead (Pb) and Cadmium (Cd) ranging from 1.21±1.6 mg/Kg (for Cd in Guava leaves) to 116.58±1.3 mg/Kg (for Mn in Mango leaves). A total of six bacterial species were isolated from both of the plants leaves (3 each). Guava endophytes were identified as Streptococcus sp, Staphylococcus albus and Staphylococcus seiuri whereas Staphylococcus aureus, Staphylococcus xylulose and Staphylococcus intermedius were from Mango leaves. The identified isolates were tested for ability to resist heavy metals in-vitro and were capable of showing different patterns of resistance to MnCl2, PbCl2 and CdCl2. All the endophytes were highly resistant to PbCl2 followed by MnCl2 but susceptible to CdCl2. The ability of plants and bacterial endophytes understudy to tolerate or resist heavy metals is a good indication of their phytoremediation potentials and thus, should be harnessed.

Khan was conducted at the Zoology lab of Ghazi University on four tree plants, i.e., neem (Azadirachta indica), mango (Mangifera indica), guava (Psidium guajava), and phalsa (Grewia asiatica). Soil samples were taken from different areas and on different months. The diversity of arthropods was analyzed through the Shannon index. The results were all significant. The total number of arthropods found in the experiment was 5151, with the following distributions: millipedes were 132 in neem, 133 in guava, 113 in mango, and 121 in phalsa; centipedes were 136 in neem, 142 in guava, 118 in mango, and 132 in phalsa; springtails were 138 in neem, 130 in guava, 120 in mango, and 134 in phalsa. There were a total of 12 different species of arthropods found. Neem (Azadirachta indica) have mites, centipede, and ants; guava (Psidium guajava) have centipedes and ants. Mango (Mangifera indica) have millipedes, centipedes, mites, springtail, and ants, and phalsa (Grewia asiatica) have mites, ants, and centipedes. The study reveals that millipedes, centipedes, springtails, and ants were found abundantly in the urban forest area of D.G. Khan, resulting in increased organic matter decomposition and appropriate distribution of nutrients through the soil having beneficial effects on the terrestrial ecosystem.
