Comparison of oregano essential oil as antibiotic or probiotic alterative in broilers̓ diet: Use of technique for order of reference by similarity to ideal solution

Antibiotic resistance is a major problem for public health worldwide, and nutritionists are searching for antibiotic alternatives. Oregano essential oil is one of the natural products that has antioxidant and antibacterial substances. In the present study the technique of ordering the preference by similarity to the ideal solution was applied to compare different feed additives regarding the biological parameters of broilers fed diets containing antibiotic, probiotic or different levels of oregano essential oil. A total of five hundred Arian broiler chicks were used in a completely randomized design with 5 treatments and 5 replicates. The experimental treatments were: 1) basal diet (control), 2) basal diet+150 ppm antibiotic Avilamycin, 3) basal diet +100 ppm probiotic Protexin, 4) basal diet+200 ppm oregano essential oil, 5) basal diet+400 ppm oregano essential oil. After obtaining data on performance, production index, heterophil to lymphocyte ratio, and ileum microflora of broilers, the multiple attribute decision making (MADM), and the technique for order of preference by similarity to ideal solution (TOPSIS) was applied. Results indicated that oregano essential oil had the potential to be considered as a natural probiotic or antibiotic replacement in broiler chickens̓ diet. Oregano essential oil improved FCR, production index, intestinal villus height/crypt depth of the broilers. Also, it decreased the bacterial count of E.Coli in the ileum content of the broilers. It conclusion using oregano essential oil at the level of 200 or 400 ppm of diet can be considered as probiotic or antibiotic natural alternative in broilers diet.

Changes in performance, egg quality and blood parameters of laying hens fed selenium and oregano oil

Context Organic and herbal additives in feed may lead to more healthy animal products. For this study, we hypothesised that an organic form of selenium and/or a herbal additive (oregano, Origanum vulgare L.) may improve performance of laying hens. Aims The study was designed to determine the effects of selenium source (SS, sodium selenite; or selenium yeast, SY), oregano essential oil (OEO) and a combination of Se and OEO on performance, egg quality and blood parameters of laying hens. Methods In total, 216 Lohmann LSL-Lite laying hens, 66 weeks of age, were assigned to feed on one of six diets: control group, receiving basal diet (BD); BD + 0.3 mg SS/kg; BD + 0.3 mg SY/kg; BD + 250 mg OEO/kg; BD + 250 mg OEO + 0.3 mg SS/kg; BD + 250 mg OEO + 0.3 mg SY/kg. For each treatment, there were six replicates of six hens each for 12 weeks. Key results Feed conversion ratio was higher (P < 0.05) with the OEO diet than in the control. The SY diet decreased egg production and the OEO diet decreased egg mass (P < 0.05), although this effect was reversed with the SY + OEO diet. This would illustrate a synergistic effect of OEO with SY. Highest and lowest yolk colour values were found with the SY and SS diets, respectively (P < 0.05). Serum uric acid was lower with diets containing Se than the control diet (P < 0.05). Glutathione peroxidase activity was highest with SY and SY + OEO diets and lowest with the OEO diet (P < 0.05). Birds fed the SS + OEO diet showed the highest total antioxidant capacity and those fed SY showed the lowest total antioxidant capacity (P < 0.05). Conclusions Based on the results of the current study, it can be concluded that SY diet have increasing effect on egg weight in laying hens. In addition, novel synergistic effects between OEO and SY diets on improving egg production, egg mass and feed conversion ratio have been found. In terms of oxidative status, the detected synergistic effects between OEO and SS diets on total antioxidant capacity and between OEO and SY diets on glutathione peroxidase (GPx) have not been reported before, that appreciate more clarifying investigations in future. Although, dietary supplemental SY individually and also with OEO increased GPx, adding SS to diet together with OEO showed more improving effect on GPx compared to the separate usage. Implications Dietary supplemental SY can improve egg weight in laying hens. In addition, adding SY to diet of laying hens individually and with OEO have beneficial effects on oxidative status of bird in terms of GPs activity, which in turn can ameliorate the unfavourable impressions of probable environmentally oxidative stress on productive performance.

Effect of Dietary Oregano and Rosemary Essential Oil Supplementation on Growth Performance and Cecal Microbiota of Broilers

In this study, the effect of dietary supplementation of oregano and rosemary essential oils (EO) on growth performance and cecal microbiota of broilers were investigated. A total of 450 1-d-old male Ross-308 broilers were divided into 5-experimental groups (10 replicates of 9 chickens): a Control (C), fed a basal diet; four treatments, which received a basal diet supplemented with oregano and rosemary EOs individually (O, 300 mg/kg oregano EO; R, 300 mg/kg rosemary EO) and combined (OR1, 150 mg/kg oregano EO + 150 mg/kg rosemary EO; OR2, 200 mg/kg oregano EO + 200 mg/kg rosemary EO). Body weight (BW), feed intake (FI), body weight gain (BWG), feed conversion ratio (FCR), and cecal microbiota (coliforms, clostridia and lactobacilli) were determined weekly, and at 42 d, respectively. BW in R (p<0.05) and OR2 (p<0.001), and BWG and FCR in OR2 (p<0.05) were significantly higher than C at 42 d, despite no difference in FI in any group during experimental period. Counts of cecal coliforms (p<0.001) and clostridia (p<0.01) decreased, and lactobacilli (p<0.001) increased substantially between C and treatment groups. Results indicated that combined oregano and rosemary EO (200 mg/kg ea) supplementation significantly increased BW and BWG, improved FCR in 1-42 d, lowered coliform and clostridial, and increased lactobacilli counts suggesting a beneficial shift in cecal microbiota.

Using the Technique for Order Preference by Similarity to Ideal Solution to Determine the Best Level of Hydrolysed Feather Powder in Broilers Diet

ABSTRACTBackgroundFeathers constitute about 5–7 percent of broilers live body weight, and they are bulky waste in poultry slaughterhouses. These wastes are a serious issue that can cause environmental pollution. On the other hand, poultry feathers can be considered as a dietary ingredient to decrease the competition between humans and poultry in consuming corn and soybeans.ObjectivesThe aim of this study was to determine the best level of hydrolysed feather powder inclusion in broilers diet by using TOPSIS.MethodsA total of five hundred Arian broiler chicks were used in a completely randomised design with 4 treatments, 5 replicates, and 25 chicks per replicate. Experimental treatments included four levels of hydrolysed feather powder as 0, 3, 6, or 9 % of the diet. The experiment lasted for 42 days. After recording parameters relating to the birds growth performance, carcass characteristics, production index, humoral immunity, heterophil to lymphocyte ratio, multiple attribute decision making (MADM), and the technique for order of preference by similarity to ideal solution (TOPSIS) were used.ResultsConsidering the closeness coefficient, hydrolysed feather at the level of 3 % had the highest coefficient (0.8038); however, hydrolysed feather at the level of 9 % had the lowest coefficient (0.7364).ConclusionsHydrolysed feather can be considered as a protein source in broilers diet that is not used as food for humans, and it will help the circular feed economy. With regard to the multi‐attribute decision‐making (MADM) method, it is recommended to use 3 % hydrolysed feather in the broilers diet.

Simple SummaryIn ruminants, fermentation control plays a crucial role in optimizing feed utilization efficiency and reducing methane emissions. Traditional approaches involving antibiotics and feed additives have been used to modify ruminal fermentation, contributing to the emergence of antibiotic resistance in humans. The use of essential oils as natural additives could potentially replace antibiotics and synthetic feed additives, promoting sustainability in livestock production. The objective of the present study was to determine the optimal dosage of a mixture of garlic and oregano essential oils as feed additives in improving ruminal fermentation. The results showed significant improvements in digestibility with the inclusion of essential oils. Garlic and oregano essential oils have the potential to modulate ruminal fermentation, improving productivity while reducing the reliance on antibiotics. These findings highlight the potential of essential oils in optimizing ruminal fermentation and their contribution to the development of sustainable animal production in ruminants.This study assessed the impact of a mixture of garlic (Allium sativum) and oregano (Origanum vulgare) essential oils (EOGOs) on in vitro dry matter digestibility (IVDMD) and in vivo apparent nutrient digestibility. Different EOGO inclusion levels were evaluated to assess the dose response and potential effects of the mixture. Three EOGO inclusion levels (0.5, 0.75, and 1 mL/kg of incubated dry matter) were evaluated in vitro, while four treatments (0.5, 0.75, and 1 mL/day of EOGO and a control group) were tested in vivo on 12 West African sheep. A randomized controlled trial was conducted using a 4 × 4 design. Blood parameters (glucose, blood urea nitrogen, and β-hydroxybutyrate) were measured to observe the effect of EOGO on the metabolism. The results showed that the inclusion of EOGO significantly enhanced IVDMD at low levels (p < 0.052) compared with the highest levels in treatments containing 0.5 and 0.75 mL/kg of EOGO dry matter. A higher intake of dry matter (DM), crude protein (CP), and neutral detergent fiber (NDF) (p < 0.05) was observed in the in vivo diets with the inclusion of EOGO. In terms of in vivo apparent digestibility, significant differences were found among treatments in the digestibility coefficients of DM, CP, and NDF. EOGO inclusion increased the digestibility of DM. CP digestibility displayed a cubic effect (p < 0.038), with the lowest values of digestibility observed at 1 mL EOGO inclusion. Additionally, NDF digestibility showed a cubic effect (p < 0.012), with the highest value obtained at 0.75 mL of EOGO inclusion. The inclusion levels above 0.75 mL EOGO showed a cubic effect, which indicates that higher concentrations of EOGO may not be beneficial for the digestibility of CP and NDF. Although no significant difference was observed in total digestible nutrients, a linear trend was observed (p < 0.059). EOGO improved the intake of DM, CP, and NDF. EOGO supplementation improved the digestibility of DM and NDF, with optimal levels observed at 0.5 mL/day. No significant effects were observed in the blood parameters. These results suggest that EOGO has the potential as an additive in ruminal nutrition to improve food digestibility and serve as an alternative to antibiotic additives. The use of EOGO potentially improves fiber digestion and may reduce the use of antibiotics in livestock production. Garlic (A. sativum) and oregano (O. vulgare) essential oils effectively modulated fiber digestibility at 0.75 mL/day. Garlic (A. sativum) and oregano (O. vulgare) essential oils have the potential to improve digestibility at low inclusion levels and serve as an alternative to antibiotic additives. The effectiveness of essential oils is greater in a mixture and at lower doses.

Oregano Essential Oil and Purple Garlic Powder Effects on Intestinal Health, Microbiota Indicators and Antimicrobial Resistance as Feed Additives in Weaning Piglets

Simple SummaryWe studied the impact of the bioactive components degradation of oregano essential oil and purple garlic powder during storage in silos, their effect on the morphometry of the jejunum and ileum and the cecal microbiota as intestinal health indicators in the piglets during the post-weaning. We also monitored antimicrobial resistance in the commensal indicator Escherichia coli. Histological parameters and intestinal microbiota were measured in 140 piglets weaned at 21 days of age. Seven dietary treatments were used: a negative control group (basal diet), a positive control group with zinc oxide (3000 mg/kg of food), two groups with oregano essential oil at 0.4% and 1.2%, respectively, two groups with purple garlic powder 0.4% and 2%, respectively, and a group with oregano essential oil with 1.2% combined with purple garlic powder with 2%. Each group of piglets received the treatment for seven weeks, from weaning, before samples were taken. Antibiotic resistance profiles were studied in 81 Escherichia coli strains from the cecal content. A progressive loss of the bioactive components of oregano essential oil and purple garlic powder was observed during the 34 days of storage. With purple garlic powder at 2% and oregano essential oil at 1.2%, their combination showed results like zinc oxide and even superior results in terms of the histological parameters studied and the counts of Escherichia coli and Lactobacillus. We observed high levels of resistance to antimicrobials of all categories. In general, the high doses of the additives studied showed the best results, obtaining levels like or higher than those offered by zinc oxide.Finding alternatives to zinc oxide is a pressing issue for the pig production sector. All categories showed high levels of resistance. Only the strains isolated from the OEO 1.2% group did not show resistance to colistin and presented the lowest resistance values. In general, high doses of the additives studied showed the best results, obtaining levels like or higher than those offered by ZnO.

Effects of novel feed additives in wheat based diets on performance, carcass and intestinal tract characteristics of quail

The present study was conducted to investigate the effects of the supplementation of an antibiotic growth promoter or novel feed additives with or without a xylanase-based enzyme complex to wheat-based diets on the growth performance, carcass yields and quality and intestinal characteristics (length of the total and small intestine, pH and viscosity of digesta, microflora) of Japanese quail. Eight hundred and forty day-old male quail chicks were randomly assigned to 14 groups of similar mean weight each of which included three replicates of 20 quail. The control group received a wheat-soyabean meal basal diet. In the treatment groups, the basal diet was supplemented with one of the following: an antibiotic, oregano essential oil, cinnamon essential oil, oregano essential oil plus cinnamon essential oil, a probiotic, a mannanoligosaccharide, and the same diets plus an enzyme. During the 35-d growth period there were no significant differences in body weight gain (BWG), feed intake (FI) or feed conversion ratio (FCR) of quail between dietary treatments. The use of oregano essential oil plus cinnamon essential oil or mannanoligosaccharide without the enzyme complex in the quail diets decreased plasma total cholesterol level compared to the control diet. The dietary supplementation of AGP, oregano essential oil, cinnamon essential oil, oregano essential oil plus cinnamon essential oil without the enzyme complex decreased plasma triglyceride levels compared with the control diet. When oregano essential oil, cinnamon essential oil, oregano oil plus cinnamon oil, a probiotic or a mannanoligosaccharide with an enzyme complex were used in a wheat based quail diet, the intestinal viscosity was significantly decreased compared to the control diet. Although the use of essential oils in combination with the enzyme complex, a probiotic and a mannanoligosaccharide with or without the enzyme complex in the wheat based diet significantly reduced the intestinal viscosity compared to the control diet, these treatments negatively decreased plasma total cholesterol and triglyceride.

Pig production depends on a health and performance balance. An approach to improve intestinal health is the oregano essential oil (OEO) supplementation within a conventional diet. Intestinal integrity regulating effects, for example gene expression, of some feed ingredients are important key factors for that balance. We hypothesized that OEO affects the expression of genes associated with pigs' intestinal integrity. In four trials, a total of 86 pigs have been used. From weaning, the 'treated' group (n=42) was additionally fed an oregano flavour additive [1500mg/kg (7.5% pure OEO)] within the basal diet. The 'control' group (n=44) was kept under identical environmental conditions, except the OEO. At age of 6months, pigs were slaughtered with an average weight of 111.1±10.9kg. In addition to automatically generated 'Fat-o-Meter' (AutoFOM) data, carcass quality factors have been measured manually. Valuable cuts of meat, such as ham and belly, were significantly reduced in the OEO group. Effects of OEO on pigs' haematologic parameters were very limited. For transcriptome analysis, the most interesting microarray expression results have been listed in a table (topTable). Selected genes were technically validated by qPCR. As a result, few significant differences in animal development and meat quality have been found between the OEO treated and the control group. Depending on OEO supplementation, we found 93 differently regulated genes in the jejunal tissue (70 up, 23 down) and 60 in the ileal tissue (48 up, 12 down). Just three genes (GRIN3B [glutamate ionotropic receptor NMDA type subunit 3B], TJP1/ZO-1 [tight junction protein ZO-1] and one uncharacterized gene) were affected by OEO both in jejunum and ileum. qPCR validation revealed AKT serine/threonine kinase 3 (AKT3), Interferon (IFN) -ε, -ω, tight junction protein (TJP1)/ZO-1 (ZO-1) to be upregulated in the jejunum and C-C motif chemokine ligand 21 (CCL21) was upregulated in the ileum of pigs that were supplemented with OEO. OEO supplementation had limited effects on pigs' performance traits. However, we were able to demonstrate that OEO alters the expression of genes associated with adaptive immune response in pigs' small intestine. These findings help to explain OEOs' beneficial impact on pigs' intestinal integrity.

Effect of dietary mannan oligosaccharide with or without oregano essential oil and hop extract supplementation on the performance and slaughter characteristics of male broilers

The effects of some alternative feed additives for antibiotic growth promoters on performance and some slaughter characteristics were examined in broilers fed wheat-soya based basal starter and finisher diets. A total of 2160 one-day-old male broiler chicks were randomly allocated to six groups with six replicate pens per treatment. The treatments were the basal diet (Control), and the basal diet supplemented with an antibiotic growth promoter (AGP); a prebiotic, mannan oligosaccharide (Bio-Mos, MOS); an essential oil of oregano (Herb-Mos Oregano, HMO); a plant extract of hop (Herb-Mos Hops, HMH) or a mixture of Herb-Mos Oregano and Herb-Mos Hops (HMOH). There were significant effects of dietary treatments on body weight and feed conversion ratio in the 0 - 21 d period, and on body weight and feed consumption in the 0 - 42 d period. The addition of all experimental additives to the diet resulted in significantly higher body weights as compared to the control treatment at both 21 and 42 days of age. Feed intakes were significantly different between the treatments at 0 - 21 d and 22 - 42 d periods, but not during the 0 - 42 d period. However, during the 0 - 21 d period the broiler chickens that received diets supplemented with AGP, MOS, HMO, HMH and HMOH had significantly better feed conversion ratios than the control group, but this pattern was not sustained during the finisher period (22 - 42 d). Mortality rate, hot carcass yield and relative weights of the small intestines, pancreas, abdominal fat and bursa of fabricius were not affected by experimental treatments. The HMH supplementation increased relative liver weight. These results showed that AGP, MOS and herbal feed additive (HMO, HMH, HMOH) supplementation to a diet provided significant advantages on broiler growth performance through a 42-d growth period. However, the combined supplementation of HMO and HMH did not exert either synergistic or additive benefits on the live performance of the broilers. These results also proved that MOS, HMO, HMH and HMOH improved broiler live performance as well as an AGP in both the starter and through the grower period. Furthermore, outstanding advantages were evidenced for the HMH treatment in particular. Therefore, the MOS, HMO, HMH and HMOH performance enhancer feed additives of natural origin may be considered as potential substitutes for AGP in broiler diets.

Effects of oregano essential oil and attapulgite on growth performance, intestinal microbiota and morphometry in broilers

Two experiments were conducted to study the effects of a blend of oregano essential oil (OEO) (as a source of natural antibacterial growth-promoting substances) and attapulgite (as a source of toxin-binder and as an antidiarrhoeal agent) on growth performance, intestinal microbiota, and intestinal morphometry in broiler chickens (Ross-308). In the first trial, the control group was fed a basal diet without antibiotic growth promoters, and the experimental group was fed the basal diet supplemented with 5% OEO (OEO) (Ecodiar ® powder at 150 g/tn) and 80% attapulgite 80% (Ultrafed ® at 6 kg/tn) blend. In the second trial, the experimental group was given the basal diet supplemented with 5% OEO (Ecodiar ® powder at 300 g/tn) and 80% attapulgite (Ultrafed ® at 3 kg/tn) blend. Intestinal microbiota was enumerated by conventional techniques with selective agar media at the end of the trial at both ileum and caecum, and intestinal morphology was assessed in the duodenum, jejunum and ileum. Results showed that in the first trial, despite the positive impact on daily gain and feed-to-gain ratio, growth performance was not affected by the blend with OEO and attapulgite. Furthermore, no effect was found on intestinal morphometry. However, the counts of lactic acid bacteria were increased significantly, and coliforms were decreased in caecal contents. In the second trial, a positive impact was noticed on daily gain and feed-to-gain ratio by the high OEO and low attapulgite blend. Dietary supplementation of OEO and attapulgite increased ileal villus height and lactic acid bacteria significantly and reduced coliforms in ileal and caecal contents compared with the control group. In conclusion, the combination of OEO at 15 mg/kg and attapulgite at 2.4 g/kg exerted a positive effect on growth performance, ileal villus height and intestinal microbiota of broilers. The Effectiveness of the Use of Oregano and Laurel Essential Oils in Chicken Feeding

The objective of this experimental study was to investigate the effects of essential oils of oregano, laurel and their combination on growth performance, intestinal microbiota and intestinal morphology as replacers of antibiotic growth promoters, as well as on the antioxidant capacity and mineral content of breast and thigh meat. A total of 256 day-old broiler chickens were randomized into 4 groups with 4 replicates. Control group received a basal corn-soybean diet, whereas the other experimental groups received the basal diet plus 25 mg/kg oregano essential oil, 2.5 mg/kg laurel essential oil or their combination, respectively. Chickens had free access to water and feed. Body weight gain and feed to gain ratio was calculated for the total fattening period and mortality was daily recorded. Intestinal microbiota was enumerated by conventional techniques with selective agar media at the end of the trial at both ileum and caecum. Also, evaluation of intestinal morphology was carried out in small intestine and caecum. At the end of the trial, birds were slaughtered, their carcasses were processed and samples of breast and thigh meat were analyzed for moisture, fat and protein content. Total phenolic content was determined in feeds and breast and thigh meat in order to assess its antioxidant capacity. Mineral content of breast and thigh meat was evaluated by ICP-MS. The results of the trial showed that the group that received oregano or the mixture of oregano and laurel presented better BW and FCR and mortality compared to control group. Bacterial counts for the Lactobacilli and Bifidobacteria were higher in the experimental groups compared to the control group at both ileum and caecum, and total coliforms were lower in caecum in the experimental groups compared to control. Higher values for villus height were found for the oregano supplemented groups compared to control or laurel ones. Oregano supplemented groups showed higher antioxidant capacity of breast and thigh meat compared to control and laurel groups, however no changes in mineral content were noted among the different groups. In conclusion, oregano essential oil alone or as a mixture with laurel essential oil can be used to improve growth performance and gut health in broiler chickens.
