Abstract

We are interested in analysing the halotolerance mechanism by comparing housekeeping genes involved in sporulation between Gram-positive, spore-forming moderately halophiles andBacillus subtilis, the model Gram-positive bacterium. One of these genes,ScoB, encoding succinyl-CoA transferase subunit B, was cloned from the moderately halophilic and alkaliphilicBacillus sp. 071 and expressed inEscherichia coli. The deduced 183 amino acids protein has an isoelectric point of 8.63. Comparative analysis of sequence showed that a conserved region was located between 47 and 55 amino acid residues as CoA-transferase active site. High percentages of cysteines and prolines were found in the ScoB protein, 2.19 and 6.01%, respectively, compared to the ScoB fromB. subtilis, which is 0.46 and 2.74%, respectively. The percentage of hydrophobic amino acid residues was slightly higher than inB. subtilis, 44.82 to 43.39%, respectively. It is the first report about the scoB gene and characteristics from Gram-positive, spore-forming, moderately halophilic and alkaliphilic bacterium.
