Abstract

This chapter explores the relationship between chemical structure, physicochemical properties and drug metabolism and how these can be controlled in the design of novel drug candidates by the medicinal chemist. The processes of absorption from the site of administration and the absorption barriers faced by a compound are discussed for several routes of administration. Tissue distribution of a compound is considered from both a physicochemical and pharmacokinetic perspective. Clearance from the body either by elimination of unchanged compound or by metabolism by a host of drug metabolising enzymes is also discussed.
