Abstract

Objective To study the changes and significance of cellular immunity in patients with different grades of glioma before and after operation. Methods 53 patients with glioma in 2019 as the research object, according to the WHO classification standard is divided into low level and high level group two groups, including low grade group (WHO grade I ~ Ⅱ) 21 cases, high grade group 32 cases (WHO I ~ Ⅱ level). The peripheral blood of the subjects was extracted in one day befor esurgery, one day after surgery, and seven days after surgery respectively. The levels of CD3+, CD4+, CD8+, and CD25+Foxp3+T lymphocytes were detected by flow cytometry, and the differences of cellular immune indexes in glioma patients of different grades and the same patients at different times were analyzed. Results The levels of CD3+, CD4+, AND CD4+/CD8+ in peripheral blood of the high grade group were all lower than those of the low grade group (54.09±4.25, 31.93±3.08, 0.93±1.06 vs 62.37±6.57, 34.88±4.43, and 1.16±0.11), but the levels of CD8+ and CD4+CD25+Foxp3+ were higher than those of the low grade group (34.23±2.48, 9.66±1.47 vs 30.16±3.75, 6.30±1.29). (p < 0.05). The levels of CD3+, CD4+, and CD4+/CD8+ in peripheral blood of patients in both high-grade and low-grade groups were all lower than those on 1d after surgery, but the levels of CD8+ and CD4+CD25+Foxp3+ were increased.(p< 0.05). The levels of CD3+, CD4+, and CD4+/CD8+ in peripheral blood of patients in the high-grade group and the low-grade group at 7 days after surgery were all higher than those at 1 day before surgery, but the levels of CD8+ and CD4+CD25+Foxp3+ were all decreased (p< 0.05). Conclusion The level of cellular immune function in peripheral blood was correlated with the grade of glioma, and the level of cellular immune suppression in peripheral blood of patients with high grade glioma was significantly lower than that of patients with low grade glioma. Surgical resection of glial tumor can significantly reduce the level of CD4+CD25+Foxp3+ regulatory T cells and reduce the degree of cellular immune suppression. T lymphocyte subsets may be one of the indexes to evaluate the degree of malignancy and prognosis of glioma patients.
