Abstract

On-line liquid chromatography/mass spectrometric (LC/MS) analysis has been extensively utilized for the structural characterization of candidate drug molecules and metabolites in biological fluids, as well as the quantitative analysis of drugs in various biological matrices. Several excellent recent primary references and reviews are available [1, 2, 3, 4, 5]. Structural characterization and quantitative analysis of drug metabolites have historically been limited by sample size, low concentrations of analytes relative to the complex biological matrix, and the presence of chemically reactive species. The use of microbore HPLC columns and volatile HPLC eluents combined with gradient elution methods has made possible the highly sensitive analysis of compounds and their polar metabolites using various ionization modes, particularly electrospray/ ionspray and continuous flow FAB methods. The use of on-line radioisotope detection coupled with LC/MS has also greatly assisted the analysis of complex metabolite profiles of radiolabeled substrates from in vivo samples [6].
