Abstract

Acid urease is immobilized onto porous glass beads with controlled pore size and packed into a small polymer column. The simple biosensing system for determination of urea comprises the immobilized enzyme column and a flow-through microcalorimetric device (enzyme-thermistor). The heat produced in the enzymatic hydrolysis of urea is calorimetrically monitored with the device. The calibration graph is linear from 0.05 to 2.0 mM urea for 1.0 ml samples. The assay takes 6 min or less. The relative standard deviation for 1.0 mM urea is 1% ( n = 10).
