Allozymic and morphological variation in the hybrid zone between Bombina bombina and Bombina variegata (Anura, Discoglossidae) in northeastern Austria

Abstract

Summary 556 specimens of fire-bellied toads (Bombina bombina and B. variegata) were collected at 17 localities in Lower Austria and the Burgenland. 18 colour pattern characters and genotypes at two marker loci were recorded for all individuals. In 212 toads from 11 populations electrophoretic variation at 12 protein loci, the index L/T and the pigmentation of testes were studied. According to measurements all specimens were diploid. Five enzyme loci (Ldh-l, Mdh-1, Idb-1, Aat, Adk) had alternate alleles fixed in species. Utilizing these genetic markers a zone of allopatric hybridization between B. bombina and B. variegate was discovered in the eastern part of the Waldviertel. Alleles from either species pre-dominate clearly in each of the populations within this zone, but most of the individuals found there are of hybrid origin. The observed pattern of differential introgression is not consistent with the dynamic-equilibrium hypothesis of maintenance of hybrid zones by gene flow, but points to stabilization of largely isolated hybrid populations. Introgression of alien alleles was also revealed in B. bombina from the Danubian Valley and in B. variegata from the Viennese Woods. Zusammenfassung Allozymatiscbe und morphologische Variation in der Hybridzone zwischen Bombina bombina und Bombina variegata (Anura, Discoglossidae) im Nordosten Osterreicbs 556 Exemplare von Rotbauch- und Gelbbauchunke (Bombina bombina and B. variegata) wurden an 17 Fundorten in Niederosterreich and im Burgenland gesammelt. Mittels dieser genetischen Marker wurde im ostlichen Waldviertel eine Zone allopatrischer Hybridisierung zwischen B. bombina and B. variegata nachgewiesen. In jeder Population dieser Zone uberwiegen die Allele jeweils einer Art deutlich, doch die meisten Individuen sind hybriden Ursprungs. Das beobachtete Muster differentieller Introgression lastt sich nicht mit der Hypothese der Aufrechterhaltung der Hybridzone durch Genflus in Einldang bringen, sondern deutet auf die Stabilisierung weitgehend isolierter Hybridpopulationen hin. Introgression von fremden Allelen wurde auch bei B. bombina aus dem Donautal und bei B. variegata aus dem Wienerwald festgestellt. 18 Zeichnungsmerkmale and der Genotyp an zwei Marker-Genorten wurden bei alien Individuen erfast. Bei 212 Unken aus 11 Populationen wurden die elektrophoretische Variation an 12 Genorten, der Index L/T and die Pigmentierung der Hoden untersucht. Auf Grund von Erythrozytenmessungen liesen sich alle Tiere als diploid einstufen. Funf Enzymloci (Ldb-1, Mdh-1, Idh-1, Aat, Adk) Wiesen artspezifisch fixierte Allele auf.
