Abstract

TMEM16F is a ubiquitously expressed bifunctional transmembrane protein for Ca2+-activated passive phospholipid scrambling and nonselective ion transduction. It plays important physiological roles in blood coagulation, bone development, immunity, inflammation and viral infection. Though recent cryo-EM studies have revealed the structures of mammalian TMEM16F in closed and intermediate states, the open, activated state remains elusive and thus leaves questions about the activation mechanism and pathways of ion and lipid transport.
