Abstract

1. 1. A 23-S rRNA component was not obtained from the mature 45-S ribosome present in Rhodopseudomonas spheroides extracts. 2. 2. The isolation of RNA in good yields in the presence of effective nuclease inhibitors is described. The procedures adopted incorporate a high degree of cell disruption. 3. 3. The high molecular weight rRNA complement of R. spheroides was found to consist of 16.25-S and 15.00-S components ( s 0 20 w values). In solvents of high ionic strength a high-molecular weight aggregate sedimenting in the 23-S region is formed which, it is suggested, accounts for other observations claiming the presence of a genuine 23-S component. 4. 4. Escherichia coli and Aerobacter aerogenes 23-S rRNA are not degraded by treatment with R. spheroides cell fractions. 5. 5. The 16.25-S and 15.00-S components are unresolved by sucrose gradient centrifugation or methylated albumin-kieselguhr chromotography but are well resolved by polyacrylamide gel electrophoresis. 6. 6. The 16.25-S and 15.00-S species are components of the 45-S ribosome and the 16.25-S species a component of the 29-S ribosome. 7. 7. The origin of the two high-molecular weight RNA components of the 45-S ribosome is discussed.
