Abstract

A proteolytic enzyme has been extracted from the latex of the plant Ficus longifolia. The optimum temperature and pH for the activity of the enzyme has been determined as 9 and 70°C respectively. It was activated by cysteine and β- mercapto ethanol. Its activity was inhibited strongly and reversibly by PCMB (para chloro mercury benzoate), which revealed that the latex protease is a cysteine protease. The protein was isolated from the crude by ion exchange chromatography. It was further purified by gel chromatography and named ficin. The molecular weight was determined to be 30 Kd by Sodium dodecyl sulphate polyacrylamide electrophoresis.
