Publisher Summary This chapter describes the assay methods, purification procedure, and properties of amino acid-activating enzymes. Amino acid-activating enzymes catalyze a reaction between ATP and various specific amino acids leading to the activation of the carboxyl group of the amino acid and the elimination of phosphate or pyrophosphate from ATP. Two general reactions have been observed: (1) inorganic pyrophosphate (Ia) or adenylic acid (Ib) are eliminated; and (2) orthophosphate is eliminated (II); X = NH20H or any other acceptor. Two general methods are used to assay for amino acid-activating enzymes. The chapter discusses the formation of amino acyl hydroxamates.
