Abstract

This paper presents capturing technique of the circulating tumor cells (CTCs)using a nucleic acid aptamer. A nucleic acid aptamer that is able to specifically bind with epithelial cell adhesion molecule (EpCAM) was used for capturing the CTCs. The specific adsorption test of a nucleic acid aptamer and cancer cell (MDA-MB-453) was carried out for verification of principle by fluorescently-labeled aptamer and fluorescently-labeled scrambled DNA. As results, the nucleic acid aptamer bind with MDA-MB-453, and the scrambled DNA did not bind with MDA-MB-453. Also, CTC capturing test was carried out using a fabricated nucleic acid aptamer-modified device. In the result of the evaluation, MDA-MB-453 was successfully captured by the nucleic acid aptamer modified on the substrate, and adsorption of healthy cell (HEK-293T) was successfully inhibited.
