Abstract
Chitosan has been developed as a polycationic carrier for pDNA in gene delivery system with promising results. On the other hand, the mechanism of the intracellular transport of pDNA/chitosan complexes remains rudimentary yet. In this study, microinjection method was employed to investigate it. The effect of pDNA concentration, post-injection time, cell cycle and stoichiometry of pDNA/chitosan complexes on expression efficiency were investigated. Stability of pDNA complexes in cytosol was also estimated.
