Abstract

Polyploid yeast strains derived from intrastrain cell-fusion were isolated with a micromanipulator from the liquid culture for fusion-regeneration of yeast protoplasts, by picking up large cells in the culture.The isolation frequencies of polyploid strains from the large cells were 6-54% on the several industrial yeast strains-wine, sake and beer yeasts. The polyploid cells were larger 1. 3-2. 3times in size, weight and DNA content per cell, and smaller in number per ml of a given medium, than the parents. In 24% glucose Yeast-extract-Peptone-Glucose medium at 20°C, their fermentations progressed slightly slower than those of the parents.
