Abstract

We improved method for transformation of Aeromonas hudrophila. The cells are gently suspended in 50mM CaCl2 containing 10mM Tris HCl(pH 8.0). after being grown with BHI broth. Then the cells were resedimented and suspended in 100mM CaCl2 containing 10mM Tris HCl (pH 8.0).After 20min at room temperature, the cells are resedimented and suspended in one-tenth the original culture volume in the same CaCl2-Tris buffer. 200μl samples are placed on ice. Following the addition of 25μl of DNA, the cells are held on ice for 20min and then heated at 42°C for 2min. Under these conditions, the transformant yield increases about 100 times more than that of Cohen's methods.
