Abstract

영양기능성이 우수한 전곡립인 현미와 흑미를 이용하여 발아 조건에 따른 전분분해효소 활성도 변화를 조사하였다. 현미의 <TEX>${\alpha}$</TEX>-amylase 활성은 3일간 발아과정 중 증가하였으며 찹쌀 현미가 멥쌀 현미에 비해 높게 나타났다. 흑미의 <TEX>${\alpha}$</TEX>-amylase 활성 역시 3일간 발아가 진행됨에 따라 계속 증가하였으며 현미와는 달리 멥쌀 흑미가 찹쌀 흑미에 비해 높았다. 현미와 흑미의 <TEX>${\beta}$</TEX>-amylase는 3일간의 발아 중 다소 미미하게 증가하였으며 찹쌀 현미와 흑미에서 높게 나타났다. 현미와 흑미의 발아과정 중 <TEX>${\alpha}$</TEX>-amylase와 <TEX>${\beta}$</TEX>-amylase 활성은 발아온도 <TEX>$30^{\circ}C$</TEX>가 <TEX>$20^{\circ}C$</TEX>에서 보다 높게 나타났다. 당화에 관여하는 효소의 활성도를 총칭하는 당화력은 찹쌀 현미와 흑미가 멥쌀에 비해 다소 높았으며 <TEX>$30^{\circ}C$</TEX> 발아온도에서 보다 높게 나타났다. 발아조건에 따른 발아 전곡립의 호화양상을 신속점도측정계로 측정한 결과 RVA 점도는 발아기간이 1-3일 경과함에 따라 감소하였으며 발아 전곡립 효소활성도가 증가함에 따른 RVA의 점도감소를 확인할 수 있었다. Enzymatic activity in germinated cereal grains is important for the saccharification of starch materials. This study was conducted to investigate the amylolytic activities of germinating brown rice and black rice that have different amylose contents. Brown rice and black rice were steeped at room temperature for 24 h and germinated at 20 and <TEX>$30^{\circ}C$</TEX> for 1, 2, and 3 days. <TEX>${\alpha}$</TEX>- and <TEX>${\beta}$</TEX>-Amylase activities in normal brown rice increased very slightly during the 3-day germination period, but the enzymatic activities were slightly higher in low-amylose (waxy type) brown rice. Diastatic power (DP), a measure of starch-saccharifying enzyme, was higher in the germinating brown rice with low amylose than in those with normal amylose content. <TEX>${\alpha}$</TEX>- and <TEX>${\beta}$</TEX>-Amylase activities in black rice increased gradually during germination, and DP of low-amylose black rice appeared to be higher than that of normal brown rice. Amylase activities in brown rice and black rice germinated at <TEX>$30^{\circ}C$</TEX> were higher than those germinated at <TEX>$20^{\circ}C$</TEX>. Compared to brown rice, the overall amylolytic activity of germinated black rice was observed to be higher than that of brown rice.
